Fos Protein

Fos protein is a short-lived transcription factor produced rapidly in response to cellular stimulation, making it a widely used marker of neuronal activity in neuroscience. Neural signals activate intracellular pathways that induce Fos expression; Fos then partners with Jun proteins to form the AP-1 complex, which binds DNA and regulates genes involved in cellular responses. Measuring Fos protein through immunohistochemistry or related methods helps researchers identify brain regions and cell populations engaged by sensory experiences, behavior, learning, or injury. These analyses connect neural activity with gene regulation and provide insight into circuit function and plasticity.

Here we present a Fluorescence Activated Cell Sorting (FACS) protocol to study molecular alterations in Fos-expressing neuronal ensembles from both fresh and frozen brain tissue. The use of frozen tissue allows FACS isolation of many brain areas over multiple sessions to maximize the use of valuable animal subjects.
