Gfp Reporter Mice

GFP reporter mice are genetically engineered animals that produce green fluorescent protein (GFP) in specific cells, making gene expression and cellular organization visible in living or preserved tissue. Researchers link the GFP gene to a cell-type-specific promoter or regulatory sequence, so cells activate fluorescence when that genetic program is active; the signal can then be detected by fluorescence microscopy or used to identify cells for analysis. In neuroscience, these mice help map neuronal populations, follow neural development, examine circuit organization, and study changes in labeled cells after injury or disease. They also support targeted cell isolation and longitudinal imaging studies.

Source: Chen, Y., et al., Measurement of Specific Mycobacterial Mistranslation Rates with Gain-of-function Reporter Systems. J. Vis. Exp. (2019)This video demonstrates a method to quantify mistranslation in Mycobacterium smegmatis using an inducible GFP and secreted mutant luciferase reporter system. It shows how luminescence resulting from mistranslation is measured and compared to GFP fluorescence to evaluate the impact of a test molecule on translational accuracy.

Stable intravital high-resolution imaging of immune cells in the liver is challenging. Here we provide a highly sensitive and reliable method to study migration and cell-cell-interactions of immune cells in mouse liver over long periods (about 6 hours) by intravital multiphoton laser scanning microscopy in combination with intensive care monitoring.
