Hippocampal Fractionation

Hippocampal fractionation is a biochemical method that separates the hippocampus into subcellular components, enabling researchers to examine where proteins, receptors, and signaling molecules are located and how they function. The procedure typically involves homogenizing hippocampal tissue in an appropriate buffer, then using sequential centrifugation steps to separate fractions such as nuclei, membranes, synaptosomes, mitochondria, and cytosol according to their size and density. Researchers analyze these fractions with techniques such as immunoblotting, enzymatic assays, or microscopy to investigate synaptic signaling, plasticity, metabolism, and neurodegenerative changes. In neuroscience, fractionation links molecular measurements to specific cellular compartments and helps clarify mechanisms underlying learning, memory, and hippocampal disease.

We provide a detailed protocol for induction of long-term potentiation in the CA1 region of the hippocampus and the subsequent isolation of nuclear enriched fractions from the tetanized area of the slice. This approach can be used to determine activity dependent nuclear protein import in cellular models of learning and memory.

Organotypic Hippocampal Slice Cultures

We describe a method to prepare organotypic hippocampal slices that can be easily adapted to other brain regions. Brain slices are laid on porous membranes and culture media is allowed to form an interface. This method preserves the gross architecture of the hippocampus for up to 2 weeks in culture.
