Hippocampus Electroporation

Hippocampus electroporation is a method for introducing DNA, RNA, or other molecular tools into cells of the hippocampus, a brain region central to learning, memory, and spatial processing. Brief electrical pulses temporarily disrupt the cell membrane, creating transient pores through which applied molecules enter hippocampal neurons or progenitor cells; the membrane then reseals, allowing gene expression or other experimental effects. In neuroscience, the technique enables targeted manipulation of hippocampal circuits, investigation of neuronal development and plasticity, and modeling of mechanisms underlying memory, neurological disorders, and potential therapeutic strategies. It is used in both developing tissue and experimental animal models.

This article describes in detail a protocol to electroporate in utero the cerebral cortex and the hippocampus at E14.5 in mice. We also show that this is a valuable method to study dendrites and spines in these two cerebral regions.
