Maternal Separation

Maternal separation is an early-life stress paradigm in which offspring are temporarily separated from their mother, allowing researchers to study how disrupted caregiving influences brain and behavioral development. During separation, changes in sensory input and maternal care can activate the hypothalamic-pituitary-adrenal axis, altering glucocorticoid signaling and stress-related neural circuits. In neuroscience, the paradigm is used primarily in animal models to investigate the developmental origins of anxiety, depression-related behaviors, learning differences, and altered social responses. Its findings help clarify how early environmental conditions shape neural plasticity and may inform research on resilience, stress regulation, and interventions for developmental and psychiatric disorders.

We are measuring perigenital mechanical sensitivity and mast cell activation in the prostate of male C57BL/6 mice that underwent an early life stress paradigm – neonatal maternal separation, in order to induce a preclinical model of chronic prostatitis/chronic pelvic pain syndrome.

Described herein is a protocol to isolate and analyze the infiltrating leukocytes of tissues at the maternal-fetal interface (uterus, decidua, and placenta) of mice. This protocol maintains the integrity of most cell surface markers and yields enough viable cells for downstream applications including flow cytometry analysis.

Assessment of Maternal Vascular Remodeling During Pregnancy in the Mouse Uterus

This protocol describes a technique to assess changes in the maternal vasculature during pregnancy in mice. Using stereological methods, remodeling of the decidual spiral arteries is assessed quantitatively and the results confirmed qualitatively using immunohistochemistry.
