Neocortical Culture

Neocortical culture is an in vitro model that maintains cells from the cerebral cortex under controlled laboratory conditions, enabling direct study of neuronal development and cortical function. Researchers typically isolate and dissociate neocortical tissue, plate the cells on a growth-supporting substrate, and provide defined nutrients that allow neurons and glial cells to survive, extend processes, and form synaptic networks. These cultures support investigations of neurite growth, synaptic activity, cellular signaling, and responses to experimental compounds using imaging, electrophysiology, and molecular analyses. As a neuroscience tool, neocortical culture complements animal and tissue-based studies by providing an accessible system for examining mechanisms of development, neural communication, and disease.

Widespread Transduction of Mouse Neocortical Neurons by Subarachnoid Injection of AAV2

A new technique for widespread delivery of adeno-associated virus that uses subarachnoid virus infusion is described. This method not only ensures widespread transduction of mouse neocortical neurons in superficial layers but also results in selective expression of the transgene in layer five pyramidal neurons, even when using a non-selective promoter.

We outline methods for the efficient and quick isolation/culture of viable microglia from the neonatal cerebral cortex and adult spinal cord. The dissection and plating of cortical microglia can be accomplished within 90 minutes, with the subsequent microglial harvest taking place ~ 10 days following the initial dissection.
