Mitochondrial Biogenesis

Mitochondrial biogenesis is the cellular process of producing new mitochondria and expanding the organelle network, enabling cells to match energy supply with metabolic demand. It involves coordinated expression of nuclear and mitochondrial genes, often regulated by PGC-1α and downstream transcription factors such as NRF1 and NRF2, which promote TFAM activity, mitochondrial DNA replication, and synthesis and import of respiratory-chain proteins. In neuroscience, this process supports the high energy requirements of neurons, synaptic activity, and axonal transport. Studying its regulation helps explain how neurons respond to stress and informs research on aging, metabolic disorders, and neurodegenerative disease.

Animal Mitochondrial Genetics

Among all the organelles in an animal cell, only mitochondria have their own independent genomes. Animal mitochondrial DNA is a double-stranded, closed-circular molecule with around 20,000 base pairs. Mitochondrial DNA is unique in that one of its two strands, the heavy, or H, -strand is guanine rich, whereas the complementary strand is cytosine rich and called the light, or L, -strand. Compared to nuclear DNA, mitochondrial DNA has a very low percentage of non-coding regions and is marked by...
