Mri Fiber Tracking

MRI fiber tracking is a neuroimaging method that estimates the course of white-matter pathways in the brain, helping researchers study structural connectivity without directly observing individual axons. It uses diffusion-weighted MRI to measure the direction-dependent movement of water, then applies mathematical models and computational algorithms to reconstruct three-dimensional fiber trajectories from voxel to voxel. In neuroscience, tractography supports investigations of brain organization, development, neurological disease, and connections between functional regions. Although the resulting pathways are estimates rather than direct anatomical images, they can help relate changes in white-matter integrity to behavior, cognition, injury, and treatment outcomes.

This video illustrates a method, using a clinical 3 T scanner, for contrast-enhanced MR imaging of the naïve mouse visual projection and for repetitive and longitudinal in vivo studies of optic nerve degeneration associated with acute optic nerve crush injury and chronic optic nerve degeneration in knock-out mice (p50KO).

We describe a general protocol for in vivo cell tracking using MRI in a mouse model with ex vivo labeled cells. A typical protocol for cell labeling, image acquisition processing and quantification is included.
