Neuroepithelium Permeability

Neuroepithelium permeability describes how readily substances cross the tightly organized epithelial layer that forms or supports nervous-system tissues, making it important for regulating the neural environment. Permeability depends on cell polarity, intercellular junctions, membrane transporters, and the physicochemical properties of each substance, which together control paracellular movement between cells and transcellular passage through them. Measuring these properties helps researchers assess barrier integrity, track the movement of drugs, nutrients, or signaling molecules, and identify changes associated with neural development, injury, inflammation, or disease. In neuroscience, these analyses support models of neural barriers and strategies for targeted delivery to nervous tissue.

An Assay for Permeability of the Zebrafish Embryonic Neuroepithelium

We describe a live whole animal quantitative measurement for permeability of the embryonic zebrafish brain. The technique analyzes the ability to retain cerebrospinal fluid and molecules of different molecular weights within the neural tube lumen and quantifies their movement out of the ventricles. This method is useful for determining differences in epithelial permeability and maturation during development and disease.

Imaging embryonic tissue in real-time is challenging over long periods of time. Here we present an assay for monitoring cellular and sub-cellular changes in chick spinal cord for long periods with high spatial and temporal resolution.
