Nmda Treatment

NMDA treatment refers to therapeutic strategies that target N-methyl-D-aspartate (NMDA) receptors, glutamate-gated ion channels that regulate synaptic signaling, learning, memory, and neuronal excitability. These interventions typically reduce or modify receptor activity: under normal conditions, glutamate and glycine binding plus membrane depolarization open the channel by relieving its magnesium block, whereas antagonists can limit excessive calcium influx and downstream excitotoxic signaling. In neuroscience, NMDA-targeting drugs such as memantine and ketamine-related therapies are studied or used in contexts including neurodegenerative disease, pain, anesthesia, and treatment-resistant depression, although their effects depend on receptor distribution, dose, and clinical setting.

Here, we describe the use of a genetically directed alkaline phosphatase (AP) labeling method to investigate the anatomical features and phenotypes of mouse retinas under NMDA-induced excitotoxicity. In conjunction with several retinal ganglion cell (RGC) subtype-specific reporter lines, we uncover novel phenotypes in dying RGCs throughout the degenerative process.
