Denatonium Benzoate

Denatonium benzoate is a synthetic quaternary ammonium salt widely recognized as an intensely bitter taste stimulus and aversive chemosensory compound. In neuroscience, it activates bitter taste receptors, including TAS2R family receptors on taste receptor cells, triggering G-protein-mediated signaling that converts chemical detection into neural activity and aversive responses. Researchers use denatonium benzoate in sensory neuroscience and behavioral assays to examine bitter-taste perception, taste coding, oral aversion, and conditioned taste-avoidance learning. Its strong, reproducible bitterness also makes it useful for testing how peripheral sensory signals influence motivation, behavior, and protective responses across experimental models.

A facile, one-pot synthesis of N-succinimidyl-4-[18F]fluorobenzoate ([18F]SFB) was developed based on a non-aqueous, three-step radiochemical process. Using microwave heating, the entire procedure can be completed in less than 30 min, or 60 min with further purification by preparative HPLC. The decay-corrected radiochemical yields (RCYs) were 35-5% (n > 30).

2016

An operationally simple procedure for the synthesis of ortho-trifluoromethoxylated aniline derivatives via a two-step sequence of O-trifluoromethylation of N-aryl-N-hydroxyacetamide followed by thermally induced intramolecular OCF3-migration is reported.
