Pharmacokinetic Safety Assessment

Pharmacokinetic Safety Assessment is the systematic evaluation of how a drug is absorbed, distributed, metabolized, and excreted, and how these processes influence safe exposure levels. By analyzing concentration-time profiles, researchers estimate parameters such as clearance, half-life, bioavailability, and brain penetration while identifying potential accumulation or dose-dependent toxicity. In neuroscience, this assessment helps determine whether candidate therapies reach the central nervous system at effective concentrations without producing unacceptable systemic or neurological effects. The findings support dose selection, study design, and interpretation of safety signals during preclinical and clinical development, improving the likelihood that treatments will be both effective and tolerable.

We exposed a microphysiological system (MPS) with intestine and liver organoids to acetaminophen (APAP). This article describes the methods for organoid production and APAP pharmacokinetic and toxicological property assessments in the MPS. It also describes the tissue functionality analyses necessary to validate the results.

Robert M. Rioux & Suprita Jharimune, Pennsylvania State University, University Park, PA Among the many hazards present in the laboratory, electrical hazards are one of the most common we must be cognizant of since most of the laboratory equipment we use requires electricity for operation. Improper handling or operation of electrical devices might lead to electric shock with the potential risk of injury or even death. Electric sparks can lead to fire or explosion (since many flammable...

Effect of Hepatic Disease on Pharmacokinetics: Pathophysiologic Assessment and Liver Function Test

2025

In clinical practice, the direct measurement of hepatic blood flow to evaluate liver function presents significant challenges due to the intricate and specialized nature of the necessary techniques. Consequently, healthcare professionals often rely on empirical estimates derived from thorough patient examinations and liver function tests to gauge liver health. Among the tools at their disposal, the Child–Pugh and MELD scoring systems stand out for their ability to categorize and assess the...

Survey Safety

Surveying near highways, rough terrain, or power lines involves significant risks. Working along highways is particularly dangerous and requires the use of warning signs and flagmen. It is safest to avoid working directly on roads and use offsets whenever possible. When highway work is unavoidable, it must follow all safety guidelines. Surveyors should wear bright clothing, such as orange reflective vests, to ensure visibility to motorists, coworkers, and hunters. In construction zones, wearing...
