Progeny Screening

Progeny screening is a method for examining offspring from controlled breeding or genetic crosses to identify inherited genotypes, phenotypes, or traits of interest. In neuroscience, researchers typically combine genotyping with observations or measurements of behavioral, anatomical, or physiological characteristics, using molecular assays and standardized phenotypic analyses to distinguish desired animals from their littermates. This process helps establish and maintain transgenic or mutant lines, verify inheritance patterns, and select subjects for studies of neural development, circuit function, and disease mechanisms. Accurate screening improves experimental consistency and reduces the risk of misclassifying animals or attributing findings to the wrong genotype.

The goal of this protocol is to alter the penetrance of lethal skeletal mutant phenotypes in zebrafish by selective breeding. Lethal mutants cannot be grown to adulthood and bred themselves, therefore this protocol describes a method for tracking and selecting penetrance through multiple generations by progeny testing.
