Puncta Analyzer

Puncta Analyzer is an image-analysis method for detecting and quantifying discrete fluorescent spots, or puncta, in microscopy images. It typically identifies puncta by applying intensity thresholds and size criteria, then measures features such as number, area, brightness, and spatial distribution within selected regions or across image channels. In neuroscience, these measurements help assess synaptic proteins, neuronal structures, receptor localization, and other molecular markers in cells and tissue. By converting fluorescence patterns into reproducible quantitative data, Puncta Analyzer supports comparisons between experimental conditions and strengthens studies of synapse formation, neuronal signaling, plasticity, and neurological disease.

Filamentous actin (F-actin) plays an important role in spinogenesis, synaptic plasticity, and synaptic stability. Quantification of F-actin puncta is therefore a useful tool to study the integrity of synaptic structures. This protocol describes the procedures of quantifying F-actin puncta labeled with Phalloidin in low-density primary cortical neuronal cultures.

Analyzing Changes in Working Capital

2025

Working capital is a key indicator of a company’s short-term financial strength and operational efficiency. Defined as current assets minus current liabilities, it measures the firm's capacity to manage day-to-day financial obligations. In financial analysis, shifts in working capital are especially significant when reviewing the cash flow statement. In the operating section of the cash flow statement, changes in working capital explain how operational decisions affect cash availability. When...

Analyzing Situations in Helping Behavior

Source: Julian Wills & Jay Van Bavel—New York University Social psychologists and personality psychologists both attempt to predict human behavior, but they focus on very different factors. Whereas personality psychologists focus on how personality traits, character, and individual differences affect behavior, social psychologists focus primarily on the power of social situations in shaping behavior. We often underestimate the role that social situations can play in determining behavior. We...
