Rat Model Construction

Rat model construction is the development of laboratory rat preparations that reproduce defined neurological, behavioral, or physiological features of human disease for controlled study. Researchers create these models through approaches such as genetic modification, targeted lesions, drug exposure, or environmental manipulation, then characterize resulting changes with behavioral, anatomical, electrophysiological, or molecular measurements. In neuroscience, carefully selected models help link cellular and circuit mechanisms to symptoms, test disease hypotheses, and evaluate potential therapies. Reproducible induction procedures, appropriate controls, and validation of construct, face, and predictive validity determine how reliably a model can support mechanistic research and preclinical translation.

Source: Svientek, S. R., et al. Fabrication of the Composite Regenerative Peripheral Nerve Interface (C-RPNI) in the Adult Rat. J. Vis. Exp. (2020).This video demonstrates the surgical construction of a composite regenerative peripheral nerve interface (C-RPNI) in a rat model of peripheral nerve injury. The procedure involves attaching a transected peroneal nerve to a muscle graft and overlaying it with a dermal skin graft to promote nerve regeneration. Over time, motor and sensory neurons...

2023

The streptozotocin-induced diabetic wound model in male SD rats is currently the most widely used model for studying wound healing in type I diabetes mellitus. This protocol describes the methods used to construct this model. It also presents and addresses potential challenges and examines the progression and angiogenic characteristics of diabetic wounds.

Creation of Reversible Cholestatic Rat Model

2011

Cholestasis is a clinical condition commonly encountered by both surgeons and gastroenterologists. Creation of a reversible cholestatic rat model can be challenging in view of the smaller size and unique hepatopancreatobiliary anatomy in rats. This video article demonstrates the creation of a reversible cholestatic model.

Here, we demonstrate fabrication of collagen-based, tissue constructs containing skeletal myoblasts. These 3-D engineered constructs may be used to replace or repair tissues in vivo. For our purposes, we have designed these as an atrioventricular electrical conduit for the repair of complete heart block[1].
