Retinal Extraction

Retinal extraction is a laboratory technique for isolating the retina from an eye or neural tissue preparation so its structure, cells, and molecular signals can be studied directly. During the procedure, researchers carefully dissect away surrounding tissues and detach the retina while minimizing mechanical damage and preserving tissue organization. This preparation supports microscopy, electrophysiology, immunostaining, gene-expression analysis, and other experiments examining photoreceptors, ganglion cells, synaptic circuits, and retinal responses. In neuroscience, retinal extraction provides a controlled model for investigating sensory processing, neurodegeneration, and how neural circuits respond to injury or treatment.

We used retinal samples from retinectomy for a transcriptomic analysis of retinal detachment. We developed a procedure that allows RNA conservation between the surgical blocks and the laboratory. We standardized a protocol to purify RNA by cesium chloride ultracentrifugation to assure that the purified RNAs are suitable for microarray analysis.
