Retrograde Anterograde Transport

Retrograde and anterograde transport are the bidirectional movements of vesicles, proteins, organelles, and signaling molecules along a neuron’s axon, enabling communication between the cell body and synaptic terminals. Anterograde transport carries cargo from the soma toward the axon terminal, primarily through kinesin motors moving along microtubules, whereas retrograde transport returns materials toward the soma through dynein. This traffic supports synaptic maintenance, membrane renewal, and neuronal signaling, while retrograde pathways can also carry neurotrophic signals and neuroinvasive pathogens. Studying these processes helps explain axonal injury, neurodegenerative disease, and how neurons maintain long-distance cellular function.

Live cell imaging of alphaherpes virus infections enables analysis of the dynamic events of directed transport and intercellular spread. Here, we present methodologies that utilize recombinant viral strains expressing fluorescent fusion proteins to facilitate visualization of viral assemblies during infection of primary neurons.

Source: Laboratories of Jonas T. Kaplan and Sarah I. Gimbel—University of Southern California Anterograde amnesia is the loss of the ability to form new memories. This can be distinguished from retrograde amnesia, which is the loss of old memories. Anterograde amnesia can result from damage to structures in the brain that are involved in the formation of new memories. Patients who have damage to the structures of the medial temporal lobe, including the hippocampus, amygdala, and the surrounding...

Source: Keefe, K. M., et al. Direct Injection of a Lentiviral Vector Highlights Multiple Motor Pathways in the Rat Spinal Cord. J. Vis. Exp. (2019).This video demonstrates the injection of a highly efficient retrograde transport (HiRet) viral vector into the rat spinal cord to trace neuronal pathways. The virus has glycoproteins for synaptic entry and a fluorescent protein coding gene. The needle is inserted into the spinal cord, and the virus suspension is administered at a controlled rate.
