Suspension Culture

Suspension culture is a cell-culture method in which cells grow and proliferate within a liquid medium rather than attaching to a solid surface, enabling controlled study of cell behavior at scale. In neuroscience, neural stem and progenitor cells can be maintained under nonadherent conditions, where growth factors such as epidermal growth factor and fibroblast growth factor support survival and promote formation of free-floating neurospheres. Researchers use these cultures to examine neural self-renewal, differentiation, and responses to experimental treatments. Suspension culture also supports more uniform sampling and expansion of neural cells, helping connect cellular mechanisms with models of development, disease, and regenerative research.

Generating an Organoid Culture from a Brain Tumor Cell Suspension

This video describes the preparation and cultivation of brain tumor organoids from single-cell suspension. Using a laminin-rich extracellular matrix as a scaffold and culture media for nutrients, cells are facilitated to multiply and self-organize within the organoid. Consequently, ex vivo organoids are generated, mimicking cellular diversity and the microenvironment of brain tumors.

The Tail Suspension Test

The tail-suspension test is validated as an experimental procedure to assess antidepressant efficacy of drug treatments in mice. Mice are suspended by their tails for six minutes and escape-related behaviors are assessed. We describe procedures used in conducting the tail suspension test.

Here we describe a robust method for the fractionation of plant plasma membranes into detergent resistant and detergent soluble membranes based on a mixture of unlabeled and in vivo fully 15N labeled Arabidopsis thaliana cell cultures. The procedure is applied for comparative proteomic studies to understand signaling processes.
