Synergistic Drug Effect

Synergistic drug effect is the enhanced biological response produced when two or more drugs act together, exceeding the effect expected from simply adding their individual actions. In neuroscience, synergy can arise when compounds influence complementary molecular targets, such as distinct receptors, signaling pathways, or stages of neurotransmission, so that one drug strengthens the impact of another. Researchers assess these interactions by comparing combined responses with responses to each drug alone, often using dose-response analyses. Understanding synergy supports the design of combination therapies for neurological and psychiatric disorders, helps clarify mechanisms of action, and guides more precise treatment strategies.

Source: Martinez, E., et al. Intracranial Pharmacotherapy and Pain Assays in Rodents. J. Vis. Exp. (2019).This video demonstrates the intracranial administration of an analgesic drug in an anesthetized rat with a pre-implanted guide cannula in the prefrontal cortex. An additional analgesic drug is co-administered intraperitoneally. Following recovery, the rat’s thermal pain response is assessed using Hargreave’s apparatus, where paw withdrawal latency is measured to evaluate the analgesic...

Divide the transwell system into five groups that undergo various treatments. The first group serves as the control and consists of a co-culture of neuroblastoma and microglia cells. In the second group, treat the co-culturer with fibrillar β-amyloid (fAβ) alone. fAβ induces damage in neuroblastoma cells. In the third group, treat neuroblastoma cells with an 808-nanometer laser, then co-culture them with untreated microglia. The laser reduces oxidative stress, improving neuronal survival. In the...

In this video, we describe a method highlighting the utility of bioluminescence assay for sensitive and rapid screening of drugs. These drugs can be tested alone or in combination to assess the synergism in their mode of action.

Synergistic Antimicrobial Effect of Membrane-Targeting Compounds on Antibiotic-Resistant Bacteria

Take suspensions of an antibiotic-resistant strain of a pathogenic bacterium.Treat one suspension with a short, membrane-disrupting antibiotic compound and the other with both the short compound and a longer, membrane-intercalating compound.Incubate both suspensions with agitation. These compounds mimic the lipid bilayer structure, enabling their spontaneous insertion into bacterial membranes. The short compound spans only part of the membrane’s thickness, creating a structural mismatch. This...
