Ventral Cuticle

Ventral cuticle is the underside of an arthropod’s external body covering, formed by epidermal cells and positioned opposite the dorsal surface. In model organisms such as Drosophila larvae, its segmental pattern of denticle belts, sensory structures, and body-wall landmarks reflects underlying developmental organization and provides a consistent orientation for anatomical analysis. Neuroscience researchers use the ventral cuticle to identify larval segments, align behavioral observations, and relate external morphology to motor circuits, neuromuscular junctions, and sensory inputs. Examining these features supports studies of nervous-system development, locomotion, and genetic mechanisms that shape neural connectivity and behavior.

Aerial plant organs are protected by the cuticle, a supramolecular biopolyester-wax assembly. We present protocols to monitor selective removal of epi- and intracuticular waxes from tomato fruit cuticles on molecular and micro scales by solid-state NMR and atomic force microscopy, respectively, and to assess the cross-linking capacity of engineered cuticular biopolyesters.

Isolation and Compositional Analysis of Plant Cuticle Lipid Polyester Monomers

Lipid polyesters constitute the structural components of two cell wall modifications, the plant cuticle and suberin-containing diffusion barriers. In this video, we describe a method to depolymerize cutin from whole delipidated leaves. The method can be applied to investigating mutants compromised in either cutin or suberin biosynthesis.
