Wheat Germ Agglutinin

Wheat germ agglutinin is a plant-derived lectin, or carbohydrate-binding protein, that recognizes specific sugars on cell-surface and extracellular glycoconjugates. It binds primarily to N-acetylglucosamine and sialic acid residues, allowing researchers to attach fluorescent, enzymatic, or neuronal tracer labels to glycosylated structures. In neuroscience, this property supports visualization of cell membranes, axonal pathways, synaptic regions, and neural connectivity in tissue samples. WGA-based labeling can reveal anatomical organization and transport within the nervous system, making it useful for microscopy, pathway tracing, and studies of how cellular glycosylation changes during development, injury, or disease.

Germ Cell Transplantation and Testis Tissue Xenografting in Mice

Protocols for germ cell transplantation and testis tissue xenografting are described. Germ cell transplantation results in donor-derived spermatogenesis in recipient testes and represents a functional reconstitution assay for identification of spermatogonial stem cells (SSCs). Testis tissue xenografting reproduces testis development and spermatogenesis of various donor species in recipient mice.

Co-cultures represent a valuable method to study the interactions between nerves and target tissues and organs. Microfluidic systems allow co-culturing ganglia and whole developing organs or tissues in different culture media, thus representing a valuable tool for the in vitro study of the crosstalk between neurons and their targets.
