Glomerular Capillaries

Glomerular capillaries are specialized networks of blood vessels within the kidney’s glomeruli that initiate urine formation by filtering plasma, making them central to renal physiology and drug handling. Their fenestrated endothelial cells, shared basement membrane, and podocyte slit diaphragms create a size- and charge-selective filtration barrier: hydrostatic pressure drives water and small solutes into Bowman’s space while retaining blood cells and most proteins. In pharmacology, glomerular filtration helps determine the renal clearance and dosing of drugs and metabolites, whereas altered permeability or reduced filtration can contribute to proteinuria, change drug exposure, and signal kidney injury.

A High-throughput Method for Measurement of Glomerular Filtration Rate in Conscious Mice

Measurement of glomerular filtration rate (GFR) is the gold standard for kidney function assessment. Here we describe a high-throughput method which allows the determination of GFR in conscious mice by using a single bolus injection, determination of fluorescein isothiocyanate (FITC)-inulin in plasma and calculation of GFR by a two-phase exponential decay model.

A technique utilizing high resolution intavital 2-photon microscopy to directly visualize and quantify gloemrular filtration in surface glomeruli. This method allows for direct determination of permeability characteristics of macromolecules in both normal and diseased states.
