Human Liver Microtissues

Human liver microtissues are three-dimensional in vitro models that organize human liver cells into compact, tissue-like structures, providing a more physiologically relevant setting for studying drug behavior than isolated cells alone. They work by preserving cell-cell interactions, extracellular matrix cues, and local nutrient and oxygen gradients that help sustain hepatocyte functions such as metabolism and secretion. In pharmacology, these models support evaluation of drug metabolism, efficacy, and liver toxicity, while enabling investigation of species-specific responses and disease-related changes under controlled experimental conditions. This information can improve candidate selection and guide safer, more predictive drug development.

This protocol describes the fabrication of elastic 3D macroporous microcryogels by integrating microfabrication with cryogelation technology. Upon loading with cells, 3D microtissues are generated, which can be readily injected in vivo to facilitate regenerative therapy or assembled into arrays for in vitro high-throughput drug screening.
