Inflammatory Cell Infiltration

Inflammatory cell infiltration is the recruitment and accumulation of immune cells within tissue following infection, injury, or other inflammatory stimuli, where it helps coordinate host defense and tissue repair. Chemokines, cytokines, and adhesion molecules activate circulating leukocytes, promote their attachment to vascular endothelium, and guide movement through the vessel wall toward signals released at the affected site. In pharmacology, measuring infiltration of neutrophils, monocytes, lymphocytes, or other cells helps characterize inflammatory disease and evaluate anti-inflammatory treatments that target mediator production, leukocyte adhesion, migration, or tissue damage. These analyses connect cellular mechanisms with drug efficacy and potential adverse effects.

The health status of the lung is reflected by the type and number of immune cells that are present in the bronchioles of the lung. We describe a bronchoalveolar lavage technique that allows the isolation and study of nonadherent cells and soluble factors from the lower respiratory tract of mice.

Isolation of Brain-infiltrating Leukocytes

A rapid method to obtain infiltrating leukocytes from the murine brain is described. This method utilizes a continuous Percoll gradient and discontinuous Ficoll gradient to select and purify the leukocyte-enriched layer. Isolated leukocytes may then be characterized by flow cytometric measurements.

This video demonstrates a method to establish a three-dimensional tumor spheroid and T cell co-culture. An agarose cast with microwells containing tumor spheroids is prepared, followed by the addition of T cells. After incubation, this setup allows studying the interaction of T cells with tumor spheroids in a controlled three-dimensional environment.

This video demonstrates a technique to quantify the relative ratio of macrophages among the total infiltrated immune cell populations in temporal artery biopsy. The immunohistochemical staining of a tissue section affected by autoimmune disease selectively stains the immune cells with specific receptors to visually differentiate them from the rest of the cells.
