Ovalbumin Specific Ige

Ovalbumin-specific IgE refers to immunoglobulin E antibodies that recognize ovalbumin, the major protein in egg white, and serves as a marker of sensitization to this allergen. In susceptible individuals, ovalbumin exposure can cross-link allergen-specific IgE bound to FcεRI receptors on mast cells and basophils, triggering mediator release and allergic inflammation. Measuring these antibodies in serum supports investigation of food-allergic responses, while controlled ovalbumin sensitization models help pharmacologists study allergy mechanisms and evaluate treatments that reduce IgE production, stabilize immune effector cells, or limit downstream inflammatory signaling. These approaches also aid assessment of therapeutic efficacy and allergic disease risk.

In vitro analysis of class switch recombination in mice is challenging due to cytophilic IgE molecules bound to Fc receptors on the surface of B cells. We describe a method for IgE detection using trypsin-mediated cleavage of surface-bound IgE prior to fixation and permeabilization for cytoplasmic fluorescence staining.

This video demonstrates a protocol for detecting serum-specific immunoglobulin E using a microfluidic cartridge-based chemiluminescence system. Serum IgEs interact with distinct allergens immobilized in the reaction zone of the cartridge, which are subsequently detected with peroxidase-conjugated anti-IgE antibodies and a chemiluminescent substrate, allowing simultaneous detection of multiple allergen-specific IgEs.

The current study describes the development and applications of a genetically engineered assay system based on the transfection of rat basophilic leukemia cells with the equine FcεRIα gene. Transfected cells express a functional receptor where the release of mediators of the allergic response can be activated by IgE and antigen.
