Starting Dose Guidance

Starting dose guidance is the process of selecting an initial medication dose that is likely to produce a beneficial pharmacologic effect while limiting toxicity. In pharmacology, it draws on pharmacokinetic and pharmacodynamic principles, including absorption, distribution, metabolism, elimination, dose-response relationships, and the drug’s therapeutic window. Clinicians and researchers adjust starting doses according to factors such as age, body size, organ function, interactions, prior exposure, and disease severity, then use monitoring and titration to refine treatment. Sound guidance supports safer clinical trials, individualized therapy, and more reliable interpretation of drug efficacy and adverse effects.

This protocol describes a bioprinting methodology using an automated robotic depositing system that incorporates etched topographical guidance cues with the precision deposition of a cell bearing hydrogel bioink. The printed cells are directly delivered to the etched features and are able to sense and orientate with them.

We describe here the use of a pH-sensitive green fluorescent protein variant, pHluorin, to study the spatio-temporal dynamics of axon guidance receptors trafficking at the cell surface. The pHluorin-tagged receptor is expressed both in cell culture and in vivo, using electroporation of the chick embryo.
