Phenobarbital Overdose

Phenobarbital overdose is a toxic exposure in which excessive amounts of this long-acting barbiturate produce potentially life-threatening depression of the central nervous system. Phenobarbital enhances gamma-aminobutyric acid type A receptor activity, increasing inhibitory signaling in the brain and causing dose-dependent sedation, impaired coordination, respiratory depression, hypotension, and coma. In pharmacology, understanding this toxicity requires relating dose, absorption, distribution, metabolism, and elimination to clinical severity, while recognizing that its prolonged half-life can sustain symptoms. Studying phenobarbital overdose supports toxicology assessment, emergency management, therapeutic drug monitoring, and safer prescribing of anticonvulsant medications.
