Reciprocal Space Mapping

Reciprocal space mapping is an X-ray diffraction technique that records diffracted intensity across a region of reciprocal space, revealing how a crystal’s lattice is arranged and distorted. During measurement, the sample and detector are rotated through a range of angles, and the resulting diffraction positions and intensities are converted into reciprocal-space coordinates related to lattice spacing and orientation. In physics and materials research, these maps help determine lattice parameters, strain, crystallographic tilt, relaxation, and epitaxial relationships in thin films, multilayers, and heterostructures. The method supports the design and characterization of engineered materials with controlled structural and electronic properties.

Reciprocal hemizygosity via sequencing (RH-seq) is a powerful new method to map the genetic basis of a trait difference between species. Pools of hemizygotes are generated by transposon mutagenesis and their fitness is tracked through competitive growth using high-throughout sequencing. Simple and reproducible procedures are described for making three structurally distinct collagen assemblies from a common commercially available Type I collagen monomer. Native type, fibrous long spacing or segmental long spacing collagen can be constructed by varying the conditions to which the 300 nm long and 1.4 nm diameter monomer building block is exposed.
