High-throughput Method

High-throughput methods are approaches for processing large numbers of samples, measurements, or computational tasks efficiently, enabling data generation and analysis at a scale that conventional workflows cannot easily support. They typically combine automation, parallel processing, standardized procedures, and structured data capture, followed by statistical quality control to distinguish reliable patterns from technical variation. In statistics, these methods support large-scale experiments, screening studies, and data-intensive research by increasing throughput, improving reproducibility, and reducing per-sample effort. Their value depends on careful experimental design, appropriate normalization, and analytical models that address dependence, missing data, and multiple comparisons.

Bicelles are lipid/amphiphile mixtures that maintain membrane proteins (MPs) within a lipid bilayer but have unique phase behavior that facilitates high-throughput screening by crystallization robots. This technique has successfully produced a number of high-resolution structures from both prokaryotic and eukaryotic sources. This video describes protocols for generating the lipidic bicelle mixture, incorporating MPs into the bicelle mixture, setting up crystallizations trials (manually as well...

GFP-fusion proteins are widely used to visualize organelles by confocal microscopy. However, screening for mutations that affect the morphology of organelles generally requires individual mutagenesis and is time consuming. Here, we demonstrate a method to simultaneously incorporate organelle-GFP markers in almost 5,000 non-essential genes in yeast.
