N Minus One

N minus one (n−1) is a statistical quantity commonly used to represent the degrees of freedom available when estimating variability from a sample. After calculating the sample mean, one constraint links the observations because their deviations from that mean must sum to zero, leaving only n−1 independent deviations; this principle motivates the n−1 denominator in sample variance and standard deviation. Using n−1 rather than n applies Bessel’s correction, reducing the tendency of sample variance to underestimate population variability. The concept supports reliable estimation, hypothesis testing, confidence intervals, and other statistical analyses based on sampled data.

We present an ECG protocol that is technically easy, inexpensive, fast, and affordable in small mice, and can be performed with enhanced sensitivity. We suggest this method as a screening approach for studying pharmacological agents, genetic modifications, and disease models in mice.

The goal of this video is to demonstrate how to perform automated DNA extraction from formalin-fixed paraffin-embedded (FFPE) reference standard cell lines and digital droplet PCR (ddPCR) analysis to detect rare mutations in a clinical setting. Detecting mutations in FFPE samples demonstrates the clinical utility of ddPCR in FFPE samples.
