Tumor Tissue Engraftment

Tumor tissue engraftment is the transplantation of tumor tissue into a living host to establish and study tumor growth under controlled experimental conditions. The implanted tissue survives when it receives nutrients and blood supply from the host, allowing tumor cells and, in some models, elements of the original tumor microenvironment to expand; immunodeficient hosts can reduce immune rejection. In statistics, researchers use engraftment rates, tumor-volume measurements, growth curves, and treatment-response data to quantify model performance and compare experimental groups. These analyses help assess biological variability, estimate treatment effects, and determine whether an engrafted model supports reproducible cancer research.

Mantle cell lymphoma (MCL) is a difficult to treat B cell disorder and it is equally difficult to establish a xenograft mouse model of primary MCL to study and develop therapeutics. Here, we describe the successful establishment of MCL xenografts in mice to help understand its underlying biology.

This video describes a method for the manual macrodissection of cancer tissue from unstained tissue specimens to obtain a specific cancer tissue. The tissue section can be processed further to obtain DNA for genome-wide studies on gastrointestinal cancers.

DCE-MRI of Orthotopic Pancreatic Tumor Xenografts: A Method to Assess Microvasculature in a Target Tumor Tissue

2023

This video describes the technique of DCE-MRI for orthotropic pancreatic tumor xenografts in mice. DCE-MRI is a non-invasive method to analyze microvasculature in a target tissue, and useful to assess vascular response in a tumor following a novel therapy. This method will assist investigators to apply DCE-MRI for orthotopic gastrointestinal cancer mouse models.
