然后，我们需要剪出适当数量的微量移液器吸头，每个孔一个，大约其长度的一半。这将作为载玻片的基础，用于创建细胞单层。接下来，将滤纸放在上面。
滤纸将用于吸收悬浮液染色中的任何多余介质。然后，塑料滑轨将作为设备的坚实支撑。然后可以用普通家用胶带将其四面密封。
最后，将整个微量移液器吸头插入设备的切割尖端。这将是悬浮染色后的细胞样品被放入的地方。最初标记载玻片中切割尖端的位置也可能有所帮助，以便更方便地识别。
我们实验室使用的免疫化学悬浮染色方案包括首先将干细胞收获到单个细胞悬液中，通常通过使用胰蛋白酶或基于 EDTA 的非酶促细胞解离缓冲液进行酶促处理。然后将细胞转移到 1 ml 2 ml 微量离心管及其标准生长培养基中。然后以 200 Gs 离心 4 分钟。
所有后续的洗涤和降速步骤将使用相同的离心速度。然后手动吸出培养基，吸液时要格外小心，以确保细胞沉淀不受干扰。然后用 1 ml PBS 洗涤两次，每次旋转并吸出。

细胞在封闭溶液中放置约 45 分钟后，离心并吸出，然后将它们悬浮在一抗溶液中。为了检测非特异性结合和背景对照，最好至少保留一个样品为阴性。未应用一抗，只是悬浮在 PBS 中。
所有样品在室温下放置约 1 小时。如果需要，可以将固定的细胞悬浮在一抗溶液中过夜，并用封闭液洗涤两次。二抗溶液可以在洗涤完成时制备。
二抗通常是对光敏感的，因此如果可能，请调暗实验室灯光并覆盖任何使用铝箔的容器，以防止荧光漂白。Antifa 试剂可以加入方案中，但在使用时应小心，因为它会导致更高的背景，例如在封闭溶液中稀释至推荐浓度的一抗。对所有样品应用一个可见度，无论洗涤完成后是否添加了初级样品。
让它在黑暗的地方（例如罐子里）静置约一个小时。用 1 ml 封闭液洗涤细胞两次。细胞现在应该已准备好使用 DPI 染料进行细胞核染色。
最初在 PBS 中制备 1 至 10， 000 稀释的 DPI 溶液，在 1 ml PBS 中洗涤细胞一次，然后复苏，在 dpi 中悬浮，让我们在室温下静置约 5 至 10 分钟。与二抗一样，DAPI 对光敏感，因此请进行必要的调整以防止荧光、漂白。用 1 ml PBS 洗涤一次，最后用 reus 洗涤。
再次悬浮在 1 ml PBS 中。然后，这应该可以用于细胞旋转装置。将最多 100 微升的单细胞溶液放入微量移液器吸头的顶部。
不过，建议用量较低，以防止不必要的溢出或滴落。之后。将细胞离心装置和平衡装置放入台式离心机中，以 1000 RPM 的速度离心 4 分钟。然后，小心地拆卸细胞旋转装置，以尽量减少细胞从载玻片上脱落的机会。
