本实验的总体目标是演示如何分离并记录初级感觉神经元，这些神经元可传递动物关节运动和位置信息，作为本体感觉信息。该过程首先解剖螯虾的腹部，暴露标本的背侧部分；第二步是将节段神经置于吸电极内，从而记录关节感受器器官的电活动。该操作流程的第三步是将关节移动到不同位置，并将运动与活动图谱相关联。最后一步是检查与感觉器官相关的解剖结构。本实验方法展示了一种记录初级本体感觉神经元活动的技术，可用于研究生物源性神经调制的影响。该方法可帮助教师讲授感觉生理学中的基本概念，例如感觉适应以及神经调质如何改变感觉感知。尽管该方法可为本体感觉和无脊椎动物的研究提供见解，但也适用于其他模式生物（如哺乳动物），以更深入地理解肌梭内的梭内肌纤维。本实验流程涵盖了如何分离、刺激并记录位于螯虾腹部肌肉中的肌感受器器官。第一步是解剖螯虾的神经以及含有肌肉感受器器官的腹部肌肉。选择一只体长为6至10厘米的螯虾（我们使用的是Procambarus clarkii），将其置于冰上2至3分钟以进行麻醉。解剖操作首先按图中A线所示位置切除螯虾头部。下一步在B处切割，以分离胸部与腹部，第三次在C处切割以打开腹部。将麻醉后的小龙虾用一只手握住螯后方。迅速从眼窝向头部中线在两侧各切割一刀，随后将小龙虾去头。制备过程中血液干燥后具有黏性，因此需立即清洁工具。将螯虾去头后，从腹面剪开胸腹部之间的连接处，剪除游泳足。如果螯虾为雄性，剪除笔状突。此时应可轻松将腹部与头胸部分离。将剪刀的一侧刀片插入腹部内，剪刀尖端朝向远离标本的方向，沿侧缘剪开。在对侧重复上述操作。用镊子的后端将肌肉和胃肠道从制备物的背侧推开，注意不要向下按压肌肉。从最后一根肋骨的侧面到另一侧面进行切割，以去除尾部的腹侧部分。将制备好的样本浸入螯虾生理盐水中，并将盛有螯虾腹部的培养皿置于解剖显微镜下。下一步是设置刺激和记录装置。该装置包括一个法拉第笼、一台显微镜、一个高强度照明器、一个显微操作器以及一个生理盐水浴。法拉第笼用于屏蔽可能干扰电生理记录的外部电场。将显微镜放置在能够俯视显微镜载物台的位置。将高强度照明器移至方便操作的位置。将吸吮电极插入显微操作器，并调整显微操作器的位置，使吸吮电极能够方便地接触盐水浴。用吸吮电极吸取盐水，直至盐水与银丝接触。将另一根导线放置在靠近吸电极尖端的位置，确保两根导线均与盐水浴接触。将交流-直流差分放大器连接至Power Lab 26T，并核对放大器上的设置。将前置放大头段连接至放大器的输入探头。将吸电极的电线连接到前置放大器上。电线连接方式为：左上端接红色，中间接绿色，底端接黑色。然后将Power Lab 2016的USB线连接到计算机。在打开计算机上的 LabChart 7 之前，请确保放大器和 PowerLab 26T 均已连接电源并开启。启动 LabChart 7，即计算机数据记录软件。LabChart 欢迎中心窗口将弹出。关闭它。点击设置。点击框体左下角的通道设置。在图表左上角，将通道数更改为一个，单击“确定”。将每秒周期数设置为约 2K。在图表右侧，将 Y 轴的电压档位设置为 500 或 200 毫伏。单击通道一。单击输入放大器。选择以下设置：单端交流耦合，如需要可选择反相以反转信号，以及抗混叠。要开始数据记录，请点击“开始”以结束数据记录。点击“停止”后，通过将一根银芯氯化银接地线一端放入浴槽，另一端连接至公共接地，从而实现浴槽的电气接地。如果接地导致浴槽在记录过程中产生电噪声，请断开浴槽的接地。下一步是暴露感兴趣的肌肉和神经。通过其螺旋状的肌纤维可识别深层内侧伸肌（DEM），而深层外侧伸肌（DEL）则具有线性排列的肌纤维。该图显示了去除了周围组织的 DEM 和 DEL 肌肉的照片，与肌肉的线描图对齐呈现。在显微镜下观察制备样本，定位背外侧肌（DEM）和背内侧长肌（DEL）。此时无法直接看到肌肉感受器器官本身，因为它们位于可见肌肉的下方。在腹部远端背外侧肌之间的第一和第二肋骨区域的中矢状部位插入两枚固定针。通过显微镜观察，找到从肌肉感受器器官传出感觉轴突的神经。寻找神经最易接近的节段。该神经呈白色，可通过移液管向标本喷洒生理盐水或轻轻吹气来观察。这会导致神经移动，从而更容易识别。找到神经后，使用显微操作器将吸吮电极直接放置在神经上方。轻轻拉动注射器，将神经吸入电极内。通过显微镜可以观察到神经被吸入电极的过程。开始实验时，请点击数据记录软件上的“开始”按钮。使用镊子轻轻将小龙虾的尾部以180度角上下移动。使用软件中的注释标记记录刺激角度，以便后续数据分析时使用。以45度角重复实验，并利用软件的注释标记记录角度的变化。再以90度角重复实验。注意在每个角度下感觉神经反应的差异。完成记录后，可直接观察肌感受器器官。该图显示了肌感受器器官的相对位置，其中 SA 和 RA 分别代表快速适应型和慢速适应型，以及 D-E-M-D-E-L 和 SEM 肌肉。右侧的照片使用亚甲蓝染色。为了在您的标本中观察肌肉感受器器官，首先需用亚甲蓝对标本进行染色。将雪茄皿中的螯虾生理盐水倒出，替换为亚甲蓝溶液，并轻轻摇晃雪茄皿数分钟。然后将多余的亚甲蓝倒入废液容器中，并将新鲜的生理盐水倒入标本上。将培养皿重新放回显微镜下，以解剖覆盖的肌肉，从而暴露肌感受器器官。用剪刀从肋骨外侧至正中矢状面进行切割，将剪刀的一侧置于肌肉下方，边向上提起边沿肌肉剪开。当切开第一和第二背外侧肌（DEL）后，将这些肌肉剥离并观察到一层称为浅层伸肌内侧肌（SEM）的薄层肌肉。肌感受器器官位于螺旋肌平行方向上的最后两条内侧肌纤维处。进行此步骤时需格外小心，以免损伤肌肉感受器器官。在螯虾的机械感受器系统中存在两种不同类型的牵张感受器。如本图所示，快相感觉单位由快速运动轴突支配，而慢相感觉单位则由慢速运动轴突支配。当张力感受器受到牵拉刺激时，它会缓慢适应刺激，并持续产生稳定的动作电位发放模式。相反，当时相性感受器受到牵拉刺激时，它会迅速适应刺激，仅产生短暂的动作电位发放模式。该记录显示了包含该感受器的腹部节段弯曲时的神经反应。首先，将该节段弯曲至45度角后放松。下一段记录显示了对90度屈曲-放松的反应。这表明，快速肌肉感受器器官对MRO丝的更大程度拉伸表现出增强的活动性。下一个录像展示了通过将尾部弯曲并保持在45度位置所引发的肌肉感受器器官活动。请注意神经活动的适应现象，表现为放电频率的降低。该实验的一个良好延伸方向是：研究神经调质是否改变了放电频率，通过在维持特定静态位置期间不同时间点测量放电频率，记录感觉反应的适应过程，并将数据填入类似此处所示的表格中，遵循本实验步骤进行操作。还可以测试其他方法，例如研究神经激素（如章鱼胺、血清素和催乳素）对初级感觉神经元化合物的细胞机制的影响，以回答更多问题。在此制备方法建立之后，研究人员描述了神经元中动作电位起始的位置，为神经生理学领域的研究者探索机械感受转导以及肢体本体感觉奠定了基础，不仅适用于无脊椎动物，也适用于脊椎动物。这一制备方法是教授神经生理学诸多基本原理的极佳教学工具。
