2. 大鼠肾内髓集合管（IMCD）原代细胞的培养
所有动物操作程序均须遵守当地及联邦法律法规。

	<li>将大鼠置于充满 CO2 的密闭箱中进行 CO2 麻醉，或使用异氟烷（U-400 麻醉机，Univentor Ltd.，马耳他；气流速度为 350–400 ml/min，异氟烷浓度为 2–2.5%）。断头放血，以便准确识别苍白的内髓质与颜色较深的外髓质之间的边界。此外，放血可减少分离所得红细胞的数量。取出肾脏，并在冰上用含庆大霉素和制霉菌素的无菌 DPBS 冲洗（见上文）。所有后续操作均需在无菌超净工作台条件下进行。</li>

精氨酸加压素（AVP）介导的水重吸收的重要性不仅体现在其与疾病的相关性上。AVP诱导携带水通道蛋白2（AQP2）的囊泡向细胞膜转运并与其融合，这一过程是一种严格依赖cAMP的胞吐作用，目前尚未被充分阐明。其他依赖cAMP的胞吐作用实例包括肾脏中肾素的分泌以及胃中H+的分泌。因此，阐明肾集合管主细胞中调控AQP2转运的分子机制，不仅有助于理解其他细胞类型中类似的分子过程，还可能为治疗与AVP介导的水重吸收障碍相关疾病的新型疗法提供方向。
阐明调控 AQP2 的机制需要获取集合管主细胞模型。目前已有多种不同的哺乳动物肾细胞系可供选择，但这些模型存在若干缺陷。在许多细胞系统中，AQP2 蛋白表达水平较低（表1；M1、HEK293、COS-7、MDCK、LLC-PK1）1-10；另一些细胞则异位过表达人源（MCD4、WT10）11,12 或大鼠（CD8）13 的 AQP2。在 MCD4 和 COS-7 细胞中，V2 受体并不表达。据我们所知，目前尚无来源于肾髓质内层集合管（即 AQP2 表达最丰富的肾区）的永生化细胞系模型，现有模型多来源于皮质集合管1,11,13-16。此类细胞模型包括广泛使用的永生化 mpkCCD（例如17）以及近期建立的 mTERT-CCD14 细胞系。这两种细胞系均可内源性表达 AQP2 和 V2R，但由于它们来源于皮质集合管，其蛋白质组很可能与髓质内层集合管（IMCD）细胞不同，例如可能表达不同的 Na+ 转运系统。
本文介绍了一种原代培养内源性表达V2R和AQP2的肾髓质集合管（IMCD）细胞的实验方案。因此，该模型最接近肾脏集合管的生理状态。由于建立该培养体系仅需常规实验室设备，其他实验室可轻松采用此方法。

原代大鼠IMCD细胞成功培养后，接种6-8天即可形成融合的单层细胞（图2）。每个60 mm培养皿中约含有6 × 106个细胞。由于培养皿预先包被了作为基底膜成分的IV型胶原蛋白18，细胞可牢固贴附于培养皿表面，因此即使经过多次彻底的洗涤操作，IMCD细胞也不会脱落。培养的细胞中最多有80%内源性表达V2R和AQP2，这些为集合管主细胞；而不表达AQP2的细胞则被认为是闰细胞，或来源于亨利袢细段、直小血管或髓质内间质细胞的非IMCD细胞。如我们先前所述，600 mosmol的培养基渗透压可防止AQP2的下调19。此外，通过在培养基中添加膜通透性cAMP类似物DBcAMP，可维持AQP2的表达。为了降低AQP2在细胞膜上的表达水平，从而促进其在细胞内的定位，IMCD细胞在实验前应于不含DBcAMP的条件下培养约24小时。若在对照条件下大部分AQP2仍定位于细胞膜，则应延长无DBcAMP培养的时间。使用标准缓冲液裂解60 mm培养皿中生长的细胞，可获得约1.5 mg蛋白质10。

本实验的总体目标是获得适合用于研究血管加压素（VP）介导的水重吸收的原代肾髓质集合管细胞，即IMCD细胞。该目标首先通过摘取大鼠肾脏来实现，第二步是从周围的外髓和皮质中分离出内髓组织。接下来，通过酶消化法处理内髓集合管以释放单个细胞。最后一步是将细胞悬浮并筛选出主细胞，然后将其接种至培养皿中。通常在接种后六到八天，细胞即可用于实验。该细胞培养系统适用于阐明精氨酸加压素（VP）介导的水重吸收的分子机制，以及可能存在的其他依赖CAP的TIC过程。此外，该系统还可用于发现治疗与VP介导水重吸收失调相关疾病的新药物候选分子及药物靶点。本实验操作将由本实验室的一名博士生，以及本实验室的两名技术人员Beata Eman和Andrea Giller进行演示。在此时间线场景之后，用胶原蛋白包被培养皿。第四型，在手术前一天即次日，首先准备培养基和酶溶液。将添加剂加入 freshly prepared DMEM 培养基中。配制酶溶液时，向DPBS滤液中加入1毫克/毫升的透明质酸酶和2.2毫克/毫升的胶原酶。将溶液灭菌后，储存于50毫升Falcon管中。每消化两份肾脏内髓质，需准备一毫升酶溶液。本方案中最困难的步骤是内髓质的解剖。为确保成功，应在解剖前将动物体内的血液排尽，以便在安乐死后更清晰地识别内髓质与外髓质之间闪亮的分界线。取出肾脏并将其置于先前准备好的无菌 DPBS 中保存。在无菌条件下操作，将肾脏从添加了庆大霉素和制霉菌素的 DPBS 溶液中转移至无菌纱布上，以去除多余的液体。接下来，使用无菌手术刀，在仍固定于纱布上的情况下，去除肾脏周围的脂肪囊。在外侧或皮质侧沿纵轴附近进行切割，注意不要完全切断，需在肾盂处保持相连，使组织仍为一整块。接下来，辨认出呈闪亮玫瑰色的外髓质和呈白色的部分内髓质。使用弯剪小心地将内髓质从周围的外髓质中分离出来。将组织置于先前配制的DPBS溶液中。将所有Meli分离并收集到冰上后，减少添加了庆大霉素和制霉菌素的DPBS溶液的体积，使其刚好浸没Meli。使用无菌剃刀片将Meli切成约5立方毫米的立方体。接下来，将无菌巴斯德吸管的尖端短暂置于火焰中，使其熔化并变圆。冷却后，用此移液器将组织悬液转移至含有新鲜无菌酶溶液的50毫升Falcon管中（酶溶液已于先前配制）。将试管置于水浴中，在37°C下持续振荡孵育1.5至2小时。此后，用移液枪反复吹打混合液数次，直至悬液呈现均匀浑浊。充分混匀后，将悬液离心五分钟。接着，弃去上清液。重悬沉淀，再次离心。重复此洗涤步骤一次。将细胞重悬于完全添加补充物的培养基中，并立即接种至培养皿或微量培养板中；接种后可直接在显微镜下评估分离的IMCD细胞、红细胞及细胞碎片。这将初步了解24小时后分离的细胞数量。用600毫渗量的DMEM洗涤细胞，并加入新鲜培养基。在接下来的一周内，应将细胞洗涤两到三次。IMCD 细胞在接种后六到八天生长至汇合时即可用于实验，如此处所示。在开始实验前，请记住将细胞在不含 DBC MP 和 INE 的培养基中孵育 24 小时。否则，水通道蛋白 2 将主要位于质膜上。原代大鼠IMCD细胞成功培养后，接种六至八天即可形成融合的单层细胞。当受到精氨酸、加压素或forskolin刺激时，AQP2从细胞内囊泡转位至质膜，如图所示。此外，如本张Western印迹所示，IMCD细胞中的AQP2表达在经加压素（VP）或forskolin处理30分钟后增加。按照本方案，可进行DNA-RNA分离、RT-PCR及免疫沉淀实验。为了分析集合管细胞中的基因及基因产物，可进行蛋白质印迹分析和免疫荧光显微镜检测。该细胞培养系统可表达内源性蛋白质，特别适用于无偏倚筛选方法。
