1. 柠檬酸盐稳定的金纳米颗粒的合成
<ol>
	<li>用王水（浓盐酸与浓硝酸按3:1体积比混合）清洗一个双颈圆底烧瓶（250 mL）和一个磁力搅拌子。 
	注意：王水溶液具有极强腐蚀性，若操作不慎，可能导致爆炸或皮肤灼伤。</li>
	<li>用去离子水彻底冲洗玻璃器皿，以去除残留的酸液。</li>
	<li>向双颈圆底烧瓶（250 mL）中加入120 mL氯金酸溶液（HAuCl4，1 mM），并在持续搅拌下加热至回流。</li>
	<li>迅速向双颈圆底烧瓶中加入12 mL柠檬酸钠三碱溶液（35 mM），并将所得混合物继续回流20分钟，以确保金盐完全还原。</li>
	<li>让纳米颗粒的胶体悬浮液（深红色）冷却至室温。</li>
	<li>使用石英比色皿（光程1 cm），在520 nm波长下通过紫外-可见分光光度法测定金纳米颗粒（AuNPs）的浓度（消光系数为2.8 × 108）。</li>
	<li>取一滴AuNPs悬浮液滴加到覆碳铜网（400目）上，在室温下干燥，然后通过透射电子显微镜（TEM）测定AuNPs的粒径。</li>
	<li>将胶体金纳米颗粒悬浮液保存于4 °C条件下。</li>
</ol>
2. 杂化膜（Au-CAM）的制备
<ol>
	<li>使用注射器装置制备嵌入金纳米颗粒的膜过滤器
	<ol>
		<li>用去离子水（10 mL）清洗由过滤装置支撑的醋酸纤维素膜（孔径：0.45 μm，直径：25 mm），重复三次。</li>
		<li>用无菌注射器（20 mL）吸取10 mL柠檬酸盐稳定的AuNPs（10 nM），缓慢加入预先清洗过的醋酸纤维素膜过滤器中（图1）。</li>
		<li>用10 mL去离子水清洗过滤装置三次，以去除未固定的AuNPs。 
		&#8203;注意：固定在醋酸纤维素膜上的AuNPs具有高度稳定性，因此Au-CAM可在常温条件下储存数周，而不会丧失其化学性质或稳定性。</li>
	</ol>
	</li>
	<li>使用真空泵制备金纳米颗粒膜过滤器
	<ol>
		<li>将醋酸纤维素膜（孔径：0.45 μm，直径：47 mm）置于烧结玻璃支撑的滤器夹具（直径：40 mm）与刻度漏斗（300 mL）之间。</li>
		<li>将烧结玻璃支撑与刻度漏斗的组合单元连接至回收瓶（500 mL）和真空泵。</li>
		<li>向刻度漏斗中加入10 mL柠檬酸盐稳定的AuNPs（10 nM），然后施加真空，直至所有AuNPs完全通过醋酸纤维素膜（约20秒）。</li>
		<li>在膜的另一侧重复相同步骤（步骤2.2.3），以在膜的两侧均固定AuNPs。</li>
		<li>在高性能条件下，使用扫描电子显微镜（SEM）分析Au-CAM表面，加速电压最高达15 kV（图2d）。 
		注意：为检测Au-CAM在高盐条件下的纳米颗粒稳定性，将复合膜浸入1.0 M NaCl溶液中2小时，随后进行目视检查以确认Au-CAM的稳定性。</li>

</ol>
3. 在批处理系统中使用 Au-CAM 吸附放射性碘
<ol>
	<li>将放射性碘（[125I]NaI，2.2 MBq）用 3 mL 纯水、1.0 M NaCl 或 10 nM NaI 稀释，并将每种溶液加入培养皿（直径 50 mm × 高 15 mm）中。 
	注意：氧化态的放射性碘可能具有挥发性，必须使用适当的铅屏蔽和铅制容器进行操作。所有放射性化学操作均应在通风良好的活性炭过滤通风橱中进行，并需使用放射性检测器监测实验过程。</li>
	<li>将通过真空过滤制备的 Au-CAM 放入放射性碘溶液中，在室温下轻轻振荡。</li>
	<li>在指定时间点（0、5、10、30、60、120 分钟）从培养皿中取出 10 μL 放射性碘溶液，使用自动 γ 计数器测定该等分试样的放射性。</li>

4. 连续流动条件下放射性碘的脱盐

		<li>将放射性碘（3.7 MBq）溶解于 50 mL 纯水、1× PBS、1.0 M NaCl、0.1 M NaOH、0.1 M HCl、10 mM CsCl、10 mM SrCl2、合成尿液或海水中。</li>
		<li>用无菌注射器（50 mL）吸取每种溶液 50 mL，并通过注射泵以约 1.5 mL/s 的流速使其流经 Au-CAM 滤膜装置（图 1）。</li>
		<li>将 5 mL 滤液转移至塑料样品瓶中，用于测定溶液中的放射性活度。</li>
		<li>使用自动 γ 计数器测定滤液中残留的放射性活度（图 4）。</li>

	</li>
	<li>Au-CAM 滤膜的可重复使用性测试

几十年来，医疗机构、科研设施和核反应堆产生了大量放射性液体废物。这些污染物常常对环境和人类健康构成切实威胁1,2,3。尤其是放射性碘，被公认为核电厂事故中最危险的元素之一。例如，关于福岛和切尔诺贝利核反应堆的环境报告表明，释放到环境中的放射性碘（包括131I（t1/2 = 8.02 天）和129I（t1/2 = 1570万年））的量超过了其他放射性核素4,5。特别地，这些放射性同位素的暴露会导致在人体甲状腺中高度摄取和富集6。此外，由于放射性碘在水中具有高溶解性，其释放可导致土壤、海水和地下水的严重污染。因此，已开展了大量研究，采用各种无机和有机吸附剂来捕获含水废物中的放射性碘7,8,9,10,11,12,13,14,15,16,17,18,19,20。尽管在先进吸附系统开发方面已投入大量努力，但在连续流动条件下具有满意性能的去污方法仍十分有限。最近，我们报道了一种新型脱盐工艺，该工艺利用由金纳米颗粒（AuNPs）构成的杂化纳米复合材料，表现出良好的去除效率、离子选择性、可持续性和可重复使用性21,22,23。其中，嵌入金纳米颗粒的醋酸纤维素膜（Au-CAM）在连续流动系统下对碘离子的脱盐效率显著高于现有吸附材料。此外，整个过程可在短时间内完成，这对于处理医疗和工业应用后产生的核废料而言是另一优势。本文的总体目标是提供Au-CAM制备的逐步操作方案24。我们还展示了一种利用工程化复合膜快速、便捷地选择性捕获放射性碘的过滤工艺。本报告中的详细操作流程将为纳米材料在环境科学研究领域的应用提供有价值的参考。

An Efficient Method for Selective Desalination of Radioactive Iodine Anions by Using Gold Nanoparticles-Embedded Membrane Filter

该方法有助于解答水处理与可持续修复领域中的关键问题，例如利用杂化纳米复合材料从水相介质中高效去除放射性碘。该技术的主要优势或突出之处在于，可通过简单的过滤过程，高效快速地去除混合盐和有机物中的放射性元素。该过程的视频演示至关重要，因为放射性液体废物的处理与操作需格外谨慎，必须采取特殊防护措施，防止放射性污染扩散，保护人员安全和环境安全。演示实验步骤的是我实验室的研究生 Ha Eun Shim。将 120 毫升氯金酸溶液加入一个已清洗的双颈圆底烧瓶中，在持续搅拌下加热至回流。迅速向双颈圆底烧瓶中加入 12 毫升柠檬酸钠三碱溶液。将所得混合物回流20分钟，以确保金盐完全还原。让纳米粒子的胶体悬浮液冷却至室温。使用石英比色皿，在520纳米波长处通过紫外-可见光谱法测定金纳米粒子的浓度。现在，将一滴金纳米颗粒悬浮液滴加到400目碳支持膜铜网上。在室温下干燥铜网，然后使用透射电子显微镜测量金纳米颗粒的尺寸。将胶体金纳米颗粒悬浮液保存在4°C条件下。使用注射器单元制备嵌入金纳米颗粒的膜滤器时，首先用去离子水清洗由滤器单元支撑的醋酸纤维素膜三次。用无菌注射器吸取10毫升柠檬酸盐稳定的金纳米颗粒，并缓慢加入预先清洗过的醋酸纤维素膜滤器中。再用10毫升去离子水清洗滤器单元三次，以去除未固定的金纳米颗粒。或者，使用真空泵制备金纳米颗粒膜过滤器时，将醋酸纤维素膜置于过滤器夹具的烧结玻璃支撑板与刻度漏斗之间。将烧结玻璃支撑板和刻度漏斗的组合单元连接至回收瓶和真空泵。向刻度漏斗中加入10毫升柠檬酸盐稳定的金纳米颗粒溶液。然后施加真空，直至所有金纳米颗粒均通过醋酸纤维素膜。在膜的另一侧重复相同操作，以在膜的两侧固定金纳米颗粒。在高压条件下，使用最高达15千伏的加速电压，通过扫描电子显微镜分析嵌入金纳米颗粒的醋酸纤维素膜（即金CAM）表面。将放射性碘用三毫升纯水、一摩尔氯化钠或十纳摩尔碘化钠稀释，然后将每种溶液加入培养皿中。操作放射性碘时应遵守标准辐射安全指南。将通过真空过滤制备的金CAM放入放射性碘溶液中。在室温下轻轻振荡。在指定时间点，从培养皿中吸取10 µL放射性碘溶液。然后，使用自动γ计数器测量该等分试样的放射性。 120 分钟后用纯化水冲洗金基 CAM。最后，使用自动伽马计数器测量膜上捕获的放射性活度。为利用金基 CAM 过滤器去除放射性碘阴离子，需将放射性碘溶解于 50 毫升的多种水溶液中。用无菌注射器吸取每种溶液50毫升，并使用注射器装置以约1.5毫升/秒的流入速率通过金质CAM滤器单元。将5毫升滤液转移至塑料小瓶中，用于测定溶液中的放射性活度。使用自动γ计数器测定滤液中残留的放射性活度。最后，按照文中的实验方案对金基CAM滤膜进行可重复使用性测试。经过30分钟孵育后，金基CAM滤膜在纯水和1摩尔氯化钠溶液中均捕获了大部分放射性碘。另一方面，在非放射性碘化钠存在的情况下，放射性的吸附被完全抑制，因为金纳米颗粒表面已被碘离子占据。将放射性碘溶液通过含有金CAM的滤膜单元。随后测定滤液中残留的放射性活度。过滤步骤获得了极高的去除效率，显著降低了放射性碘的浓度。特别是，金基CAM在高浓度无机盐（如钠、铯和锶）存在下仍能保持良好的脱盐性能，未受到抑制。此外，金基CAM可重复用于合成尿液和海水中的放射性碘脱盐。尽管该方法可为从多种水相介质中修复放射性废物提供见解，但通过在金纳米颗粒表面引入金属螯合剂和无机复合物，该方法还可拓展应用于其他放射性核素及有毒重金属的去除。通过该方案，可进一步探索并设计自动化的污染水处理系统，以实现本研究在大规模工业应用中的实际推广。请务必注意，操作放射性材料可能具有极高危险性，在执行该方案时，必须采取充分的预防措施，例如在通风良好的通风橱中操作、使用个人防护装备，并尽量减少与有害物质的接触。
