两天后，检查细胞。大多数细胞应该看起来健康和明亮。然而，如果超过三分之一的细胞出现黑暗，倾斜烧瓶，用20毫升的低bFGF介质替换20毫升的介质，并辅以每毫升20毫微克的bFGF。
第三天，取出一半的介质，用15毫升的HESC介质代替每毫升bFGF10毫微克。第五天，用15毫升的神经感应介质替换一半的介质。之后，每隔一天，用神经感应介质替换一半的介质。
培养三周后，如果需要，在最终浓度为 1 倍时，将 100 倍青霉素-链霉素添加到介质中。每隔一天刷新一半的媒体。在这个协议中，形成的大脑器官看起来明亮和大小相似，没有黑暗垂死的细胞在这些集群的中心。
细胞逐渐从bFGF中分离开。在第五天，他们被放入神经诱导介质，他们在整个文化时期都留在这种介质中。虽然器官随着时间的推移变得更大，因此更暗，神经玫瑰花环状结构扩大，这表明神经分化的开始，并包含胚胎神经管的特征。
为了更深入地研究细胞内的基因表达，进行了qPCR分析。谷氨酸运输器VLGUT1在两个半星期内表达，在五周时增加，并在五个月的培养中保持一致。前脑标记Foxg1在低水平表达，直到五个星期的文化。
