注意： 本方案中使用经R848（TLR7/8激动剂）处理的CAL-1 pDC细胞系。该方案已应用于其他多种人源和小鼠细胞类型，包括RAW 264.7（小鼠巨噬细胞系）、THP-1（人单核细胞系）、BJAB（人B细胞系）、Ramos（人B细胞系）和MUTZ-3（人树突状细胞系）23。
1. CAL-1 细胞的刺激
<ol>
	<li>在无菌条件下，将 CAL-1 细胞培养于 T75 培养瓶中，置于 37 °C、5% CO2 培养箱中，使用含 5% 胎牛血清（FBS）、25 mM HEPES 和 1× 巯基乙醇的 RPMI 1640 培养基（即完全 RPMI 1640 培养基），每瓶加入 20−25 mL。</li>
	<li>将细胞转移至 50 mL 圆底离心管中。 
	注意： CAL-1 细胞为非贴壁细胞。对于贴壁细胞类型，可采用标准胰酶消化法收集细胞。</li>
	<li>在室温（RT）下以 200 × g 离心 5 分钟，弃上清液，将细胞沉淀重悬于 8 mL 完全 RPMI 1640 培养基中，制成均一的单细胞悬液。</li>
	<li>使用血细胞计数板对细胞进行计数。以每孔 1 × 106 个细胞的密度，将细胞接种于 6 孔板中，每孔加入 4 mL 预热的完全 RPMI 1640 培养基。于 37 °C、5% CO2 培养箱中孵育 20−24 小时，使细胞融合度达到 90%−95%（相当于约 1.5 × 106 个细胞）。</li>
	<li>通过向 6 孔板每孔加入 4 µL 的 1 mg/mL R848（终浓度为 1 µg/mL）刺激细胞。同时设置未刺激对照孔，即不含 R848 处理的细胞孔。</li>
	<li>轻轻左右摇动培养板，确保 R848 均匀分散。随后将细胞置于 37 °C、5% CO2 培养箱中孵育 2−16 小时。</li>
</ol>
2. 细胞蛋白的提取
<ol>
	<li>将6孔板中的细胞悬液转移至5 mL离心管中。</li>
	<li>在室温下以200 × g 离心5分钟，弃去上清液，并将细胞沉淀重悬于1 mL磷酸盐缓冲液（PBS）中，以获得均一的单细胞悬液。</li>
	<li>将细胞悬液转移至1.5 mL离心管中。</li>
	<li>在4 °C下以12,000 × g 短暂离心0.5−1分钟，小心吸除上清液。</li>
	<li>配制裂解缓冲液：在250 mL去离子水（ddH2O）中加入6.25 mL 1 M Tris-HCl（pH 7.4，终浓度为25 mM）、7.5 mL 5 M NaCl（终浓度为150 mM）、0.5 mL 0.5 M EDTA（终浓度为1 mM）、2.5 mL NP-40（终浓度为1%）和7.5 mL甘油（终浓度为5%）。使用前立即加入100倍蛋白酶抑制剂单次使用混合液，使终浓度为1倍。将配制好的裂解缓冲液置于冰上保存。 
	注意：不含蛋白酶抑制剂的裂解缓冲液可在4 °C下保存。</li>
	<li>将细胞沉淀用30 µL预冷的裂解缓冲液重悬，并通过反复吹打混匀。</li>
	<li>在冰上孵育15−20分钟。</li>
	<li>在4 °C下以12,000 × g 离心15−20分钟，澄清裂解物。将上清液转移至新的预冷1.5 mL离心管中。所有操作过程中均需将提取物保持在冰上。 
	注意：细胞裂解物可在-20 °C或-80 °C下保存。切勿煮沸样品。</li>
	<li>使用Bradford试剂测定蛋白浓度。</li>
</ol>
3. 通过非变性PAGE分析IRF5二聚化
<ol>
	<li>配制上层（-）和下层（+）电泳缓冲液。上层缓冲液由含0.3%脱氧胆酸钠（NaDOC）的1×非变性PAGE电泳缓冲液组成，下层缓冲液仅使用1×非变性PAGE电泳缓冲液。 
	注意：每次新电泳运行均需配制新鲜的上层缓冲液。</li>
	<li>用清水彻底冲洗一块3%−12%的非变性PAGE胶，注意不要破坏加样孔。将凝胶放入小型电泳槽中并拔去梳子。在4 °C冷藏环境或冰上以150 V预电泳30分钟。 
	注意：预电泳可去除过量的氨和过硫酸盐离子，这些离子可能干扰凝胶电泳过程。</li>
	<li>在预电泳期间，将保存在冰上的细胞蛋白与4×非变性上样缓冲液混合，制备上样样品。</li>
	<li>预电泳结束后，每样品上样10−15 µg蛋白，最终体积为10−15 µL。 
	注意：蛋白上样量过高可能导致条带拖尾。</li>
	<li>以85 V电泳30分钟，随后以150 V电泳2小时。 
	注意：考虑到不同实验室所用设备和细胞系的差异，可能需要对蛋白样品浓度、电压和电泳时间进行微调以优化本实验方案。降低预电泳和电泳电压并延长电泳时间，有助于提高二聚体的分辨能力和结果的一致性。</li>
	<li>将凝胶在SDS电泳缓冲液（25 mM Tris pH 8.3，250 mM 甘氨酸，0.1% SDS）中室温浸泡30分钟。 
	注意：此步骤无需振荡。由于脱氧胆酸盐（DOC）存在时可能导致转膜效率低下，条带强度可能与上样蛋白量不成正比，这一现象主要影响IRF5的单体形式。在转膜前将凝胶浸泡于SDS电泳缓冲液可解决此问题。凝胶较为脆弱，操作时应从凝胶底部（即浓度较高的一端）小心处理。</li>
</ol>
4. IRF5的免疫印迹分析
<ol>
	<li>将聚偏二氟乙烯（PDVF）膜浸入甲醇中约 5 分钟以激活。</li>
	<li>在膜的一角剪一个切口以标记其方向。按照制造商说明书中的顺序小心组装转膜夹心结构，确保无气泡残留。</li>
	<li>将转膜夹板放入转膜槽中，在冰上以 20 V 恒压转移 1 小时。 
	注意： 后续所有孵育和洗涤步骤均需在摇床上进行。</li>
	<li>转膜完成后，用塑料镊子取出膜，并在封闭缓冲液（TBS）中于室温封闭 45 分钟。 
	注意： 含 5% BSA 的 TBS 缓冲液也可用作封闭缓冲液。</li>
	<li>将膜与表 1中所列的一抗孵育。用 1× TBST 洗涤缓冲液（20 mM Tris，pH 7.0，150 mM NaCl 和 0.1% Tween 20）在摇床上洗涤 3 分钟，重复洗涤 2 次。</li>

干扰素调节因子5（IRF5）是一种重要的转录调控蛋白，在调节免疫应答中发挥关键作用，特别是在促炎性细胞因子和I型干扰素（IFNs）的释放过程中1,2,3。IRF5的异常调控是多种自身免疫性疾病的重要诱因，这一点已通过IRF5基因位点上的多种多态性得到证实，这些多态性与系统性红斑狼疮、多发性硬化症、类风湿关节炎等疾病密切相关4,5,6,7,8,9,10。因此，在生理相关的细胞环境中，建立一种可靠的内源性IRF5活化状态检测方法，对于深入理解IRF5的调控通路及其下游效应至关重要。
IRF5在单核细胞、树突状细胞（DCs）、B细胞和巨噬细胞中组成性表达1,11。与其他IRF家族转录因子类似，IRF5在静息状态下位于细胞质中。激活后，IRF5被磷酸化并形成同源二聚体，随后转位至细胞核，结合I型干扰素（IFNs）和促炎性细胞因子编码基因的特异性调控元件，最终诱导这些基因的表达1,2,11,12,13。IRF5调控多种Toll样受体（TLRs）下游的先天免疫应答，例如定位于内体、依赖MyD88进行信号传导的TLR7、TLR8和TLR91,11,14。这些TLRs主要识别感染特征性的外源核酸，如单链RNA（ssRNA）和非甲基化的CpG DNA15,16,17,18。研究表明，IRF5可调控针对细菌、病毒和真菌感染的免疫应答19,20,21。鉴于IRF5在免疫系统中具有重要且多样的作用，增强或抑制IRF5活性可能为开发新型治疗药物提供一条新途径22。因此，建立一种监测内源性IRF5激活状态的实验方案，对于深入研究在不同类型细胞中调控IRF5活性的通路与机制至关重要。

天然聚丙烯酰胺凝胶电泳（native PAGE）是一种广泛用于分析蛋白质复合物的方法31,32。与十二烷基硫酸钠聚丙烯酰胺凝胶电泳（SDS-PAGE）不同，native PAGE 根据蛋白质的形状、大小和电荷进行分离，同时在不使蛋白质变性的前提下保持其天然结构31,33,34,35。本方案利用了 native PAGE 的这些特性，可检测 IRF5 的单体和二聚体形式。该方法在检测早期激活事件方面尤为重要，因为目前尚无适用于检测内源性磷酸化 IRF5 的商业化抗体。此前已有若干研究采用 native PAGE 来评估 IRF5 的二聚化，但其中大多数研究依赖于外源表位标签标记的 IRF5 过表达，以分析其激活状态2,13,24,36,37。本文提供了一种逐步操作的实验方案，通过改良的 native PAGE 技术，在人浆细胞样树突状细胞（pDC）系中分析内源性 IRF5 的二聚化，而 IRF5 的活性在该细胞系中已被证实对其功能至关重要1,38,39,40。该技术同样已应用于其他细胞系23。

天然聚丙烯酰胺凝胶电泳免疫印迹分析内源性IRF5二聚化

本方案采用一种广泛应用的技术，结合可商业获取的试剂和工具，用于评估刺激早期内源性IRF5的激活情况。该方法可轻松应用于其他细胞背景下IRF5生物学功能的研究。尽管非变性PAGE是一种常用技术，但要获得清晰、稳定、可解释且可重复的结果在技术上具有挑战性。使用商用的电泳运行缓冲液和凝胶系统有助于最大限度地减少实验变异性。关注实验细节至关重要，而丰富的经验是获得良好结果的关键。该改良的非变性PAGE实验方案包含多个步骤，其中许多细微之处对实验成功至关重要。视觉演示可以展示这些对研究人员有益的细节。在37摄氏度和5%二氧化碳条件下，使用完全RPMI 1640培养基在T75培养瓶中维持CAL-1细胞培养。当准备刺激细胞时，将细胞转移至50毫升锥形管中，并在200 × g下离心5分钟。弃去上清液，用培养基重悬沉淀，以获得均一的单细胞悬液。随后使用血细胞计数板对细胞进行计数，并以每孔1,000,000个细胞的密度将细胞接种至六孔板中，每孔加入4毫升预热的培养基。将细胞培养20至24小时，使细胞融合度达到90%至95%。次日，通过每孔加入4微升浓度为1毫克/毫升的R848刺激细胞，同时设置对照孔，仅含细胞而不加R848处理。轻轻左右摇动培养板，使 R848 均匀分散，然后将细胞在 37 摄氏度、5% 二氧化碳条件下孵育 2 至 16 小时。孵育结束后，将细胞悬液从培养板转移至 5 毫升离心管中。在 200 × g 条件下离心 5 分钟，弃去上清液，并将沉淀重悬于 1 毫升 PBS 中。将细胞悬液转移至1.5毫升离心管中，在4摄氏度下以12,000 × g离心30至60秒，然后小心吸去上清液。根据论文所述方法配制裂解缓冲液，并置于冰上预冷备用。用30微升预冷的裂解缓冲液重悬细胞沉淀，上下吹打混匀。然后将离心管置于冰上孵育15至20分钟。在4°C条件下，以12,000 × g离心15至20分钟，澄清裂解液，并将上清液转移至预冷的1.5毫升离心管中，确保提取物始终置于冰上。随后，使用Bradford试剂测定蛋白质浓度。根据文稿说明配制上层和下层电泳缓冲液，并用水彻底冲洗一块3-12%的天然PAGE凝胶，注意不要破坏加样孔。操作脱氧胆酸钠时需格外小心。处理该化学品时必须佩戴个人防护装备，如实验服、护目镜和口罩。随后将凝胶安装至Mini Gel Tank中，取出梳子，加入已配制的上层和下层电泳缓冲液，并在4摄氏度的冷室或冰上以150伏电压预电泳30分钟。同时，在冰上将细胞蛋白与4×天然样品缓冲液混合，以准备上样。预电泳完成后，向每个加样孔中加入10至15微克蛋白，在85伏电压下电泳30分钟，随后在150伏电压下继续电泳2小时。然后将凝胶置于SDS电泳缓冲液中，室温下浸泡30分钟。将聚偏二氟乙烯膜浸入甲醇中约五分钟以激活。在膜的一角剪去一小角以标记其方向，然后根据制造商说明书组装转膜夹心结构。将转膜夹板放入转膜槽中，在冰上以20伏电压转膜1小时。然后用塑料镊子从胶片盒中取出膜，并在室温下于封闭缓冲液中于摇床上封闭45分钟。接着，将膜与一抗在4摄氏度孵育过夜，或在室温孵育2小时，随后在摇床上用1×TBST洗涤缓冲液洗涤3分钟。然后，在室温下用1×TBST洗涤缓冲液将膜与二抗再次孵育45分钟，并重复洗涤步骤。然后使用适当的凝胶成像系统扫描印迹。采用本实验方案，对经R848刺激或未刺激的CAL-1细胞进行免疫印迹分析。在未刺激的CAL-1细胞中，IRF5在非变性PAGE上表现为单一条带，对应其单体形式。对于刺激后的细胞，IRF5 单体水平降低，而二聚体水平升高。在转染了不同构建体的IRF5过表达293T细胞上，使用抗IRF5抗体进行了免疫印迹实验。未转染的对照组中未检测到IRF5，证明了抗IRF5抗体的特异性。仅在过表达IRF5的293T细胞中检测到一条对应于IRF5单体的条带。当共转染编码IRF5激活蛋白的质粒时，出现了一条迁移较慢、对应于IRF5二聚体形式的条带。然而，与RIG-I相关的蛋白NMDA5并不能诱导IRF5发生二聚化。使用 Bis-Tris 梯度凝胶至关重要，这可能是由于该凝胶电泳系统的特定 pH 值和化学组成能够分离 IRF5 的单体和二聚体形式。此外，使用非变性天然样品缓冲液裂解并保存细胞裂解物可保留蛋白质的天然结构。本实验中，我们使用了一种市售的、专为天然 PAGE 酸设计的缓冲液。另一种与此方法高度互补的技术是利用 ImageStream 成像流式细胞术系统来评估 IRF5 的核转位，这是 IRF5 二聚化后激活过程中的关键事件。与本方案结合使用时，该方法可作为验证 IRF5 激活过程中各步骤的另一种检测手段。作为炎症反应的关键调控因子，IRF5 在感染与免疫、自身免疫性疾病、癌症以及多种影响人类健康的重要疾病中发挥着重要作用。目前也有致力于开发靶向IRF5及相关转录因子的治疗手段。本方案将帮助不同领域的研究人员理解并深入探究IRF5的生物学特性。
