由于其具有特异性和典型的行为参数，且这些参数可通过本方案轻松量化，因此收缩行为是一种强大的行为表型，使研究人员能够开展机制性研究，以解析感觉通路及行为的神经元调控机制25,28。此外，已有研究表明，在神经毒理学研究中，收缩行为可作为敏感的终点指标，用于检测化学物质对神经系统发育和功能的不利影响22,24,25,30。由于多种不同的感觉通路似乎通过不同机制汇聚并诱导收缩行为28，因此与其他涡虫行为不同，研究人员可利用多种但特定的刺激来解析不同的神经回路，并研究不同信号如何整合以产生收缩表型。
重要的是，不同物种之间存在差异，同一种化学物质可能在一种涡虫物种中引发收缩运动，而在另一种涡虫中则引发不同的行为反应。例如，我们发现花生四烯酸乙醇胺（anandamide）可诱导日本三角涡虫（Dugesia japonica）产生收缩运动，但在地中海涡虫（Schmidtea mediterranea）中却诱导出蠕动运动28。这一实例凸显了能够可靠区分不同运动步态的重要性，因为这些步态是不同分子机制的表型表现。然而，仅依靠定性的观察数据难以区分收缩运动与蠕动运动，因为这两种运动均由肌肉驱动，且在表现特征上具有相似性7,28。因此，有必要通过纤毛成像或定量行为学研究来区分这两种步态，这些方法可根据特征性参数进行判别7,28。由于纤毛成像在实验操作上具有挑战性，且需要高倍复合显微镜和高速相机等专用设备7,28，其在研究者中的普及程度不如定量行为分析。

Planarian Scrunching as a Quantitative Behavioral Readout for Noxious Stimuli Sensing

淡水涡虫通常依靠腹侧纤毛的推动而平滑滑行。然而，某些环境条件可诱导其产生由肌肉驱动的运动方式：收缩运动（peristalsis）或蜷缩运动（scrunching）。虽然收缩运动是由纤毛功能缺陷引起的，但蜷缩运动与纤毛功能无关，是涡虫对特定刺激的一种特异性反应，这些刺激包括截肢、有害温度、极端pH值以及乙醇。因此，这两种由肌肉驱动的运动方式在机制上是不同的。然而，它们在定性上可能难以区分。本文提供了一种利用多种物理和化学刺激诱导蜷缩运动的实验方案，并详细描述了如何使用免费软件对蜷缩运动进行定量表征，从而将其与收缩运动和滑行运动区分开来。由于蜷缩运动是一种普遍存在于各类涡虫中的运动模式，尽管存在物种特异性的差异，本方案在适当考虑物种差异的前提下，可广泛适用于所有涡虫物种。为验证这一点，我们比较了行为学研究中最常用的两种涡虫——Dugesia japonica 和 Schmidtea mediterranea——对同一组物理和化学刺激的反应。此外，由于蜷缩运动具有高度特异性，本方案还可与RNA干扰技术或药理学处理相结合，用于解析参与该行为的分子靶点和神经回路，从而为伤害感受和神经肌肉通讯的重要机制提供深入见解。

眼外近紫外光感知在 S. mediterranea 涡虫的TRPA1依赖性机制已被提出与H相关2O2 释放17. 因为 H2O2 暴露会诱导TRPA1依赖性的收缩反应 S. mediterranea 和 D. japonica 涡虫28，可使用第2.1.4节中的步骤来检测近紫外光照射是否在这两个物种中均诱导了scrunching现象。尽管 D. japonica 涡虫在暴露于近紫外光时会收缩（10/10） S. mediterranea 涡虫如前所述表现出尾部变细（7/10）17 或无反应（3/10）（图4A,4B）。按照第1.4节所述，对挤压参数进行量化，以 D. japonica 表现出至少3次连续直线收缩...

该方案通过将定量行为分析与分子和化学干扰相结合，使研究人员能够阐明涡虫的行为及其行为背后的机制。该技术易于掌握，适用于所有技术水平的研究人员，因为其无需使用高级仪器或专用软件即可获得自由移动涡虫的定量行为读数。收缩行为可用于研究涡虫的异常反应，并可作为检测外源性物质和疾病引起神经系统功能紊乱的敏感终点指标。首先将一个可调光的LED面板放置在平坦的表面上，以提供均匀的白色背景，并可作为可调节的光源以获得适当的对比度。将直径100毫米的培养皿实验区域置于LED面板上。将相机安装在实验区域上方的铁环架上，并根据需要调整其位置、高度和焦距，使整个实验区域位于视野中央且清晰对焦。用约 25 毫升适当的暴露培养基填充实验区域以达到最大体积，打开 LED 面板，并关闭所有可能影响录制质量的其他光源。使用移液管将一只涡虫放置于实验区域中央。当测试化学溶液时，应尽量减少涡虫所携带的涡虫培养水，以避免显著改变化学物质的浓度。开始以 Fiji 原生格式记录图像序列数据。对于滑行实验，记录一到两分钟的滑行行为；对于收缩或蠕动实验，记录足够长时间以捕获至少三次连续的、沿直线发生的振荡。实验完成后，停止记录。如果涡虫在未满足终止条件的情况下到达了实验区域的边界，则用移液器将涡虫移回实验区域的中心。在 Fiji 中打开某项实验的原始图像序列，将其转换为 8 位，并使用图像堆栈底部的箭头工具或滑块浏览图像序列。为了提取特定时间段和感兴趣区域，使用矩形工具绘制一个包含涡虫完整运动路径的感兴趣区域。右键单击图像堆栈并选择“复制”，勾选“复制堆栈”框。输入感兴趣序列的第一帧和最后一帧，然后单击“确定”。对于滑行实验，提取一段涡虫滑行的时期，期间涡虫移动距离至少为其体长的两倍；对于收缩或蠕动实验，则提取相应的运动阶段。提取涡虫沿直线完成至少三次连续且完整身体摆动的实例。对复制的图像序列应用阈值，以实现图像二值化并从背景中提取涡虫；调节滑动条，使整个涡虫被红色高亮显示。具体数值取决于成像质量。保留暗背景、叠加直方图的选项，并取消勾选重置范围。滚动浏览图像堆栈以确保阈值范围合适，然后点击应用。在“将堆栈转换为二值图像”窗口中，将方法设置为默认，背景设置为亮。取消勾选此窗口中的所有复选框，然后点击确定。将出现一幅二值化图像，显示白色背景上的黑色涡虫。确保在图像序列的每一帧中，整个涡虫均清晰可见。通过点击“分析”并选择“设置测量”来设定测量参数。勾选面积、质心、堆栈位置和拟合椭圆的选项框，然后点击确定。选择打开的图像堆栈，然后依次选择“分析”和“分析颗粒”。在“分析颗粒”窗口中，选择“显示”和“掩膜”，以打开一个新堆栈，显示使用所选参数检测到的所有对象。通过输入涡虫的近似面积来设置大小过滤器，以去除不需要的噪声。然后勾选“显示结果”和“清除结果”复选框，点击“确定”。使用面板底部的滑块浏览掩膜图像堆栈，确保没有噪声或不含涡虫的帧。在结果窗口中，通过“文件”和“另存为”保存数据，将文件名添加 CSV 扩展名，以逗号分隔值格式保存数据。图像堆栈的数据保存后，关闭相应的图像堆栈结果窗口和掩膜窗口。为了通过有害温度诱导涡虫收缩，将涡虫培养用水置于玻璃烧杯中，在加热板上加热至65摄氏度。将一只涡虫放置在观察区域中央，等待其调整为正立方向并开始滑行后，开始记录。使用P200移液器，缓慢且稳定地将100微升预热的涡虫培养水滴加到涡虫咽后部的尾端，以诱导其发生收缩反应。一旦收缩停止，即停止记录。将涡虫放入恢复容器中，若需继续进行实验，则更换培养皿中的培养液为新鲜的室温涡虫用水。若需通过截肢诱导收缩，将一条涡虫转移至行为检测区域中央，等待涡虫调整为直立方向并开始滑行。然后开始录像，使用洁净的剃刀片将涡虫截断，确保每次实验中的切割位置保持一致。当前段部分停止收缩后，停止录像。将两部分取出，放入单独的容器中，使其再生七天。待截肢的涡虫完成再生后，可将其重新放回原容器中。本实验方案用于检测近紫外光照射是否会在 S.mediterranea 和 D.japonica 涡虫中诱导卷曲行为。结果显示，D.japonica 涡虫在近紫外光照射下会发生卷曲，而 S.mediterranea 涡虫则表现为尾部变细或无任何反应。对表现出至少三次连续直线收缩的日本三角涡虫（D.japonica）的收缩参数进行量化，揭示了该物种特有的收缩参数特征。相比之下，在小鼠中暴露于250微摩尔肉桂醛（一种未知的TRPA1激动剂）会引起收缩行为，且在地中海涡虫（S.mediterranea）中也观察到类似现象。而日本三角涡虫则表现出蛇形运动与振荡运动的混合模式，其间被滑行或剧烈的头部转向所打断。对至少出现三次连续振荡的样本进行量化分析，结果显示四个参数中有三个显著低于日本三角涡虫收缩行为的预期值，表明所观察到的振荡运动并非收缩行为。RNAi 验证了地中海涡虫（S.mediterranea）在肉桂醛暴露下收缩反应的特异性。在涡虫水中暴露180秒内，所有对照RNAi组涡虫均出现收缩，而SMTRPA-1 RNAi组涡虫则无一发生收缩，表明地中海涡虫在肉桂醛作用下的收缩反应依赖于SMTRPA-1。在操作动物时保持一致性和规范性至关重要，这有助于减少噪声并确保行为测量结果的可重复性。本项目可进一步扩展，以包括体型分析，从而能够识别和量化本研究中未捕捉到的其他涡虫行为。
