固定舞台插入的玻璃底盘，并添加额外的E3介质，以保持胚胎在一夜之间湿润。使用建模粘土将塑料盖密封在盘子上，并提高与菜肴接触的目标。在显微镜软件的标题下，单击添加以保存第一个样品的 XYZ 信息，以便延时，并选择具有强荧光和最佳安装的示例。
在定位选项卡下，搜索下一个示例并单击位置并添加以选择所示示的示例。当所有示例都已选中时，突出显示第一个位置并单击移动到。在持续扫描时，将光学囊泡排列在框架内，相对于光学囊泡和大脑，在前部和相交区域留出充足的空间。

设置激光功率和增益后，停止扫描并单击更新以更新位置信息。要移动到下一个位置，请单击位置二。一旦第一个和最后一个 Z 切片被定义为演示，打开时间系列标题并将周期数设置为 300，时间间隔设置为 2.5 分钟。
在查看设置以确保所有内容都完成后，单击开始延时并确认第一轮成像捕获正确，然后才允许延时运行一夜。直接注射到单个细胞是统一标签和强力荧光的必要条件。当胚胎沉浸于高糖中时，胚胎应在整个盘子中均匀地间隔。
如果胚胎的分期正确、荧光充足且安装充分，它们将在延时期间保持在成像框架中，从而能够对整个器官进行成像。不沿正点轴方向的样本（如水平或对角线的样本）会随着延时而从成像框架中长出，无法用于进一步分析。过度旋转的胚胎会导致更后延。
旋转不足的胚胎只能观察到最前组织。此外，安装在框架方向上的样品更有可能产生成功的延时。这种对腹腔侧的偏向为腹腔方向的组织生长提供了额外的空间，从而产生最佳的延时。
当不符合这些标准时，延时将不再捕获组织发展的整个 3D 体积，无法用于进一步分析。这些信息丰富的数据集可以通过多种方式进行分析，包括具有细胞速度和轨迹量化的 4D 细胞跟踪、细胞和组织体积测量以及 3 和 4D 可视化。
