由于新鲜猪角膜是食品工业的副产品，机构动物护理和使用委员会无需批准其用于研究。与用于研究的人类角膜不同，猪角膜不存在生物危害问题，未使用的眼部组织可作为普通垃圾处理。本研究所用的试剂和设备列于材料表中。
1. 器官培养的准备工作
<ol>
	<li>使用前，在最低必需培养基（MEM）中添加青霉素-链霉素作为补充物。</li>
	<li>通过向含有5 mM葡萄糖（相当于90 mg/dL）的补充MEM培养基中加入3.6 g D-葡萄糖，配制高葡萄糖培养基，使终浓度达到25 mM（相当于450 mg/dL），以模拟糖尿病患者的高血糖状态。</li>
	<li>通过向20 mL含5 mM或25 mM葡萄糖的补充MEM培养基中加入0.2 g琼脂糖，并在微波炉中加热至琼脂糖完全溶解，制备1%琼脂糖溶液。</li>
	<li>将含琼脂糖的溶液转移至维持在48 °C的水浴中。</li>
	<li>实验前，对所有实验试剂进行高压灭菌处理，包括蒸馏水、PBS、手术器械（止血钳、镊子、手术刀柄、剪刀和打孔器）以及烧杯、纸巾、无绒擦拭布、手套和移液器吸头。将硅胶模具、刀片和刀片架浸泡于70%酒精中30分钟，并用无菌PBS洗涤三次。</li>
</ol>
2. 角膜培养用猪眼球的制备
<ol>
	<li>从当地屠宰场获取猪眼球，并将其置于冰上装有湿润腔的容器中运送到实验室。 
	注意：牛眼也可采用类似方式使用。然而，牛眼和角膜与人眼/角膜的差异更为显著。例如，人和猪角膜具有5-7层上皮细胞，而牛角膜约有20层13,14。此外，牛眼在角膜器官培养过程中更易受到污染；因此，统计分析需要更多的牛眼样本。</li>
	<li>将猪眼球放入含有灭菌PBS的1 L烧杯中。 
	注意：猪眼球在屠宰后1小时内采集，并在约2小时内运送至实验室。总体而言，眼球在4小时内使用（步骤2.3）。</li>
	<li>用镊子夹住一个眼球&#160;，在无菌培养皿中用剪刀去除眼外组织。</li>
	<li>用蒸馏水冲洗眼球一次，再用PBS冲洗两次。</li>
	<li>用聚维酮-碘消毒液冲洗眼球10秒，随后用灭菌PBS洗涤三次。</li>
	<li>将眼球置于含20 µg/mL庆大霉素的PBS中孵育30分钟，并用PBS冲洗两次。</li>
</ol>
3. 上皮损伤
<ol>
	<li>用无菌无尘布固定眼球，并用6 mm角膜环钻标记角膜中心。</li>
	<li>用小型手术刀或刀角钝化的软剃须刀片轻轻刮除环钻标记区域内的上皮细胞，彻底清除细胞碎片但保留基底膜完整，再用棉签清洁创面。 
	注意：收集了上皮细胞的手术刀&#160;可转移至置于&#160;冰上的微量离心管中。细胞可立即裂解，或储存于-20 °C冰箱中作为对照使用。</li>
</ol>
4. 角膜器官培养和 离体 高血糖建模
<ol>
	<li>用手术刀和剪刀沿角膜-巩膜缘切开眼球，将角膜取出，并在含有磷酸盐缓冲液（PBS，pH 7.4）的无菌1000 mL烧杯中冲洗。</li>
	<li>将切除的角膜倒置放入由无毒液态模具硅胶制成的无菌模具中。 
	注意：将无毒硅胶模具液倒入50 mL组织培养/试管架的孔中，使用30 mL超速离心管底部压入液体中，形成半圆形模具。</li>
	<li>用保持在 48 °C 含 1% 琼脂糖的 MEM 填充角膜内皮腔，并让混合物凝固&#160;室温</li>
	<li>将角膜翻转并转移至35 mm培养皿中，然后将2 mL含或不含待测试剂的MEM培养基滴加到角膜中央表面，覆盖角膜缘结膜区域，使上皮暴露于空气中（图1).</li>
	<li>将培养皿置于湿润的5% CO₂培养箱中2 在37 °C培养箱中孵育，并每天更换含或不含待测试剂的新鲜MEM培养基，持续2天。</li>
	<li>建立一个 离体 通过向MEM中添加L-葡萄糖以达到25 mM总葡萄糖浓度，用于建立高血糖模型，并用于1%琼脂糖凝胶制备及作为培养基。</li>
</ol>
5. 角膜功能评估
<ol>
	<li>通过Richardson染色法对受损角膜进行染色以标记残留的伤口区域，并拍摄照片（图2和图3），从而确定上皮伤口愈合速率。使用Image-J或Photoshop软件（直方图分析）对伤口大小进行定量分析。</li>

Evaluating Therapeutic and Chemical Toxicity Using Organ-Cultured Porcine Corneas and Epithelial Wound Healing

白内障手术是全球最常开展的手术之一，而滴眼液在术后护理中起着至关重要的作用。白内障手术后使用滴眼液有助于预防眼部感染、炎症和黄斑水肿等并发症。非甾体抗炎药（NSAID，NS）滴眼液，包括酮咯酸、溴芬酸和奈帕芬酸，通常用于白内障手术前、手术中及术后的缓解眼部疼痛和肿胀。长期使用含有不同浓度防腐剂（如苯扎氯铵）的此类滴眼液，可能对角膜健康产生不良影响22。

我们正在研究糖尿病伤口愈合过程中角膜的固有免疫。具体而言，我们的目标是理解为何糖尿病患者更容易受到细菌感染，以及高血糖导致伤口愈合延迟的原因。我们发现，IL-1β 与 IL-1 受体拮抗剂之间，以及 TGF-β1 与 TGF-β3 之间的平衡对角膜伤口愈合具有重要作用。这可能为糖尿病性角膜病变提供新的治疗方法。我们的模型模拟了糖尿病患者中人类伤口愈合延迟的情况，使我们能够研究细胞程序性死亡通路并开发靶向治疗策略。首先，取置于1升烧杯中的猪眼。用镊子夹住眼球，在无菌培养皿中用剪刀去除眼外组织。进行上皮损伤时，使用无菌无尘纸巾固定眼球，并用6毫米角膜环钻标记角膜中心位置。然后，使用小型解剖刀或尖角钝化的软质剃须刀片，轻轻刮除角膜环钻标记圈内的上皮细胞。清除所有细胞碎片，同时确保基底膜保持完整。随后，用棉签清洁伤口区域。使用手术刀和剪刀沿角膜巩膜缘切开眼球进行解剖。然后，将角膜放入含有磷酸盐缓冲液（PBS）的无菌500毫升烧杯中冲洗。接着，将切除的角膜倒置放入无菌模具中。用含有1%琼脂糖（保持在48摄氏度）的最低必需培养基（MEM）填充角膜内皮腔。然后将角膜倒置并转移至35毫米培养皿中。以滴加方式在中央角膜表面加入2毫升MEM，可含或不含待测试剂，确保覆盖角膜缘结膜区域，同时使上皮暴露于空气中。然后，将培养皿置于37°C、含5%二氧化碳的湿化培养箱中。在损伤后40小时，用Richardson染色液对受损的培养角膜进行染色。染色后，在拍照前用PBS洗涤角膜。然后，使用相同尺寸的环钻标记原始伤口，并用小型手术刀刮取标记圈内的上皮细胞。收集刮下的细胞并转移至预冷的离心管中。与使用溴芬酸 0.09% 和酮咯酸 0.45% 处理的角膜相比，使用奈帕芬酸 0.1% 处理的角膜剩余伤口面积显著更大。在高葡萄糖条件下，用 LL-37 在 0.2 和 0.5 微克/毫升浓度下处理的角膜与未处理的角膜相比，伤口愈合显著加快。
