然后将细胞以 500G 离心 8 分钟,弃去上清液,然后将沉淀重新悬浮于 500 μL 细胞悬液中。使用自动细胞计数仪对 10 μL 等分试样的细胞进行计数。用 75% 乙醇彻底清洗 Hamilton 注射器 3 到 4 次。

然后用超纯水冲洗注射器 3 到 4 次,以确保去除所有乙醇残留物。对斑马鱼进行麻醉后,通过运动减少和反射反应丧失来确认麻醉深度。将鱼的背侧朝上,头部向左放在干净的湿纸巾上,以稳定注射。

牢牢抓住 Hamilton 注射器,将针头与尾静脉成大约 45 度角。对于 Casper 斑马鱼,通过半透明身体瞄准容器的可见部分。轻轻地将针头插入容器中,并施用从 mpo:EGFP 转基因斑马鱼中分离的 2 微升全肾骨髓细胞。

注射后,用棉签轻轻按压注射部位 10 秒以控制出血。在显微镜下观察注射的斑马鱼,以确认血管内的 GFP 信号。让注射的斑马鱼在新鲜消毒的鱼水中恢复 10 分钟。

然后将鱼转移到静态水箱中。静脉注射方法的成功率最高,43 条鱼中有 41 条鱼可见 GFP 信号。而眼眶后方法在 37 条鱼中只有 3 条检测到 GFP,而心内方法显示 40 条鱼中有 11 条成功。

静脉注射方法导致注射鱼的存活率更高,尤其是与眼眶后和心内方法相比,细胞剂量较高。早在移植后第 3 天即可在静脉注射鱼中检测到 GFP 信号,而眼眶后和心内方法显示 GFP 信号出现延迟。到第 17 天,静脉注射的鱼在肾中显示出最突出的 GFP 信号。

肾骨髓流式细胞术显示,静脉注射的鱼中约有 18% 的 GFP 阳性细胞,是 5 个细胞的 10 倍,与供体肾骨髓中的 GFP 水平对齐。相比之下,心内和眼眶后方法分别显示 5% 和 2% 的 GFP 阳性细胞。
