What are Proteins?

Das Protein ist eine Kette von Aminos&#xE4;uren, die aus Amin- und Carboxylgruppen zusammengesetzt ist.
 Gemeinsam mit verschiedenen Seitengruppen, bezeichnet als R, an einem Karbonkern gebunden sind.
 Aminos&#xE4;uren sind durch kovalente Peptidbindungen verbunden.
 Die sich wiederholende Reihe von Atomen entlang des Kerns wird das Polypeptidr&#xFC;ckgrat genannt.
 Die Sequens beginnt am N-Terminus mit einer freien Aminogruppe NH3 plus und endet mit dem C-Terminus, eine freie Carboxylgruppe COO minus.
 Jedes Protein ist einzigartig, abh&#xE4;ngig von der Anzahl der Aminos&#xE4;uren und deren Seitenketten.
 Das Schilddr&#xFC;sen-Releasing-Hormon hat 234 Aminos&#xE4;uren, wohingegen das verbundene elastische Muskelprotein &#xFC;ber vierunddrei&#xDF;ig Tausend hat.
 Au&#xDF;erdem gibt es zwanzig verschiedene Seitenketten, die in eine von vier Gruppen fallen.
 Negative polare Aminos&#xE4;uren, wie die Asparagin- und Glutamins&#xE4;uren, haben Seitenketten mit einer Carboxylgruppe.
 Deswegen agieren diese wie S&#xE4;ure mit neutralem pH-Wert.
 Positive polare Aminos&#xE4;uren wirken wie Basen und haben eine Aminogruppe in ihrer Seitenkette.
 Mehrere Aminos&#xE4;uren sind polar, aber ungeladen.
 Sie neigen dazu hydrophil zu sein und formen den &#xE4;u&#xDF;eren Abschnitt eines gefalteten Proteins, wo sie frei mit dem aquatischen Umfeld interagieren k&#xF6;nnen.
 Und schlie&#xDF;lich eine gro&#xDF;e Aminos&#xE4;urengruppe enth&#xE4;lt unpolare Seitenketten.
 Sie k&#xF6;nnen einfach sein, wie bei Glyzin oder komplex sein, wie Prolin und Tryptophan.
 Unpolare Seitenketten machen im Allgemeinen Aminos&#xE4;uren hydrophob, und bilden den Kern vieler Proteine.

 Proteine sind Ketten von Aminos&#xE4;uren, die durch Peptidbindungen verbunden und in einer 3-dimensionalen Struktur gefaltet sind. Die Seitenketten der einzelnen Aminos&#xE4;urereste bestimmen die Wechselwirkungen zwischen den Aminos&#xE4;ureresten und damit letztendlich auch die Faltung des Proteins. Je nach L&#xE4;nge und Komplexit&#xE4;t der Struktur klassifiziert man Ketten von Aminos&#xE4;ureresten als Oligopeptide, Polypeptide oder Proteine.
 
 <h4>Die Bausteine der Proteine sind die Seitengruppen der Aminos&#xE4;urereste</h4>
 
 Eine Aminos&#xE4;ure ist ein Molek&#xFC;l, das eine Carboxyl-(COOH) und eine Aminogruppe (&#x2013;NH2) enth&#xE4;lt, die an das gleiche Kohlenstoffatom gebunden sind. Dieses Kohlenstoffatom bezeichnet man als &#x237A;-Kohlenstoff. Die Identit&#xE4;t der Aminos&#xE4;ure wird durch ihre Seitenkette oder ihren Seitenrest, der oft als R-Gruppe bezeichnet wird, bestimmt. Die einfachste Aminos&#xE4;ure ist Glycin. Die R-Gruppe des Glycins ist ein einzelnes Wasserstoffatom. Andere Aminos&#xE4;uren tragen komplexere Seitenketten. Die Seitenkette bestimmt dabei die chemischen Eigenschaften der Aminos&#xE4;ure. Sie kann beispielsweise Wasser anziehen oder absto&#xDF;en (hydrophil oder hydrophob sein). Des Weiteren kann sie auch eine negative Ladung tragen (sauer sein) oder Wasserstoffbr&#xFC;ckenbindungen bilden (polar sein).
 
 Von allen bekannten Aminos&#xE4;uren werden nur 21 zur Proteinbildung in Eukaryonten verwendet. Der genetische Code kodiert sogar nur 20 davon. Aminos&#xE4;uren werden mit einem Drei-Buchstaben-Code (z.B. Gly, Val, Pro) oder einem Ein-Buchstaben-Code (z.B. G, V, P) abgek&#xFC;rzt. Die lineare Kette der Aminos&#xE4;urereste bildet das R&#xFC;ckgrat des Proteins. Die freie Aminogruppe an einem Ende wird als N-Terminus bezeichnet, w&#xE4;hrend die freie Carboxylgruppe am anderen Ende den C-Terminus bildet. Die chemischen Eigenschaften der Seitenketten bestimmen in hohem Ma&#xDF;e die Endstruktur des Proteins, da sie miteinander und auch mit polaren Wassermolek&#xFC;len interagieren.
 
 Um ein Polypeptid zu bilden, werden Aminos&#xE4;uren durch Peptidbindungen miteinander verbunden. Peptidbindungen werden zwischen der Aminogruppe (&#x2013;NH2 Gruppe) einer Aminos&#xE4;ure und der Carboxylgruppe (&#x2013;COOH) der benachbarten Aminos&#xE4;ure gebildet. Proteine werden durch eine Kondensationsreaktion unter Wasserabspaltung gebildet. Bei der Verkn&#xFC;pfung von zwei Aminos&#xE4;uren wird jeweils ein Wassermolek&#xFC;l gebildet. Die resultierende kovalente Bindung ist eine Peptidbindung.
 
 <h4>Der pH-Wert der Umgebung bestimmt die chemische Funktion der Aminos&#xE4;uren</h4>
 
 Aminos&#xE4;uren haben sowohl eine basische als auch eine saure Gruppe. Sie k&#xF6;nnen daher als Base (Empf&#xE4;nger von Wasserstoffionen) oder als S&#xE4;ure (Spender von Wasserstoffionen) fungieren. Die chemische Eigenschaft h&#xE4;ngt vom pH-Wert des umgebenden Mediums ab. Bei niedrigem pH-Wert (z.B. pH-Wert von 2) sind sowohl die Carboxyl-, als auch die Aminogruppe protoniert (&#x2013;NH3,&#x2013;COOH). So wirkt die Aminos&#xE4;ure als Base. Bei einem alkalischen pH-Wert (z.B. ein pH-Wert von13) sind sowohl die Carboxyl- sowie die Aminogruppen deprotoniert (&#x2013;NH2, &#x2013;COO-) und die Aminos&#xE4;ure fungiert als S&#xE4;ure. Bei einem neutralen pH-Wert (d.h. in den meisten physiologischen Umgebungen, ~pH7,4) wird die Aminogruppe protoniert (&#x2013;NH3) und die Carboxylgruppe deprotoniert (&#x2013;COO-). Dadurch entsteht ein Zwitterion, welches ein Molek&#xFC;l mit sowohl positiver als auch negativer Ladung ist. Diese chemischen Eigenschaften bei physiologischem pH-Wert sind wesentlich f&#xFC;r die Bildung von Wasserstoffbr&#xFC;ckenbindungen, die wiederum zur Bildung komplexerer Proteinstrukturen beitragen.
 
 <h4>Die L&#xE4;nge, strukturelle Komplexit&#xE4;t und Funktionalit&#xE4;t unterscheiden ein Polypeptid von einem Protein</h4>
 
 Polypeptide sind Ketten von Aminos&#xE4;uren. Polypeptide mit weniger als 20 Aminos&#xE4;uren werden auch als Oligopeptide oder einfach Peptide genannt. Eine Polypeptidkette wird als Protein bezeichnet, wenn sie zu einer dreidimensionalen Struktur gefaltet ist, die es erm&#xF6;glicht spezifische zellul&#xE4;re Funktion auszu&#xFC;ben.
 
 <h4>Proteine sind die wesentlichen Bausteine des Lebens</h4>
 
 Proteine bilden zusammen mit Kohlenhydraten, Nukleins&#xE4;uren und Lipiden grundlegende Bausteine des Lebens. Sie weisen eine enorme Vielfalt in ihrer Zusammensetzung und somit auch in ihren Funktionen auf. Einige Funktionen erm&#xF6;glichen zum Beispiel die Zellstruktur (z.B. in Form von Kollagen), die Bewegung (z.B. Aktin und Myosin in den Muskeln), das Katalysieren von Reaktionen (Enzyme), oder den Transport von Molek&#xFC;len durch die Zellmembran sowie die Abwehr von Eindringlingen in Wirbeltieren (Antik&#xF6;rper).

EXPLANATION: Discussions of DNA in earlier grades have been limited to discussions of genes and the role of genes in the appearance and activities of an organism. Biology 1 represents a student's introduction to a molecular model of a DNA as well as the organization of DNA into genes and genes subsequently into chromosomes. Students should address interactions between genes in proteins which regulate both the shape and reproduction of DNA molecules. It is important to note that not all segments of DNA are transcribed for proteins. Portions of DNA are involved in regulating the expression of genes or affect the structure of the chromosomes (replication origin, centromeres, and telomeres).
