Protein Organization

<p>Wenn Aminos&#xE4;uren w&#xE4;hrend der Translation aus dem Ribosom austreten, bilden sie die erste prim&#xE4;re Proteinstruktur, eine verkettete Sequenz, deren L&#xE4;nge variieren kann.
  Diese Peptidkette wird durch kovalente Bindungen zwischen den Amin- und Carboxylenden von zwei Aminos&#xE4;uren zusammengehalten.
  Wenn sich das Peptid verl&#xE4;ngert, bilden sich Wasserstoffbr&#xFC;cken.
 </p><p>Einige Aminos&#xE4;uren binden sich an benachbarte, wodurch sich der Strang verk&#xFC;rzt und kr&#xFC;mmt, wodurch eine helikale Struktur entsteht, die als Alpha-Helix bezeichnet wird.
  Andere haften seitlich und werden als Beta-Sheets bezeichnet.

 <p>Proteine sind grundlegende Bausteine des Lebens und erf&#xFC;llen viele verschiedene Funktionen in der Zelle. Sie werden aus Aminos&#xE4;uren gebildet. Die Reihenfolge der Aminos&#xE4;uren wird als Prim&#xE4;rstruktur eines Proteins bezeichnet. Lokale Wechselwirkungen einzelner Aminos&#xE4;uren f&#xFC;hren dazu, dass sich diese lineare Kette zu Sekund&#xE4;rstrukturen faltet. Interaktionen entfernter Aminos&#xE4;uren f&#xFC;hren zu einer weiteren Faltung des Proteins, welche man Terti&#xE4;rstrukturen nennt. Der Aufbau mehrfach gefalteter Ketten (Untereinheiten) wird als Quart&#xE4;rstruktur bezeichnet.</p>

 <p>Aminos&#xE4;uren, die in einer Kette miteinander verbunden sind, werden als Polypeptide bezeichnet. Diese Aminos&#xE4;uren sind durch ihre Amino (&#x2013;NH<sub>3</sub>) und Carboxyl (&#x2013;COOH) Gruppen miteinander verbunden, welche Peptidbindungen bilden. Diese Ketten an verkn&#xFC;pften Kohlenstoff- und Stickstoffatomen bilden das Grundger&#xFC;st eines Proteins. Die Aminos&#xE4;ureseitenketten stehen dabei senkrecht ab. Die Reihenfolge der Aminos&#xE4;urereste in der Polypeptidkette ist die Prim&#xE4;rstruktur.</p>
 
 <h4>Wasserstoffbindungen zwischen naheliegenden Aminos&#xE4;ureresten bestimmen die Sekund&#xE4;rstruktur</h4>
 
 <p>Die Amino- und Carboxylgruppen des Proteinger&#xFC;stes k&#xF6;nnen Wasserstoffbr&#xFC;ckenbindungen bilden. Wenn mehrere nah beieinander liegende Aminos&#xE4;urereste Wasserstoffbr&#xFC;ckenbindungen bilden, k&#xF6;nnen sich lokale Strukturen wie Alpha-Helices und Beta-Faltbl&#xE4;tter bilden. Dies sind h&#xE4;ufig lokalisierte Strukturen und bestimmen somit die sogenannte Sekund&#xE4;rstruktur eines Proteins.</p>
 
 <h4>Die Wechselwirkungen zwischen entfernten Seitenketten bestimmen die Terti&#xE4;rstruktur</h4>
 
 <p>Die Terti&#xE4;rstruktur eines Proteins beschreibt die drei-dimensionale Anordnung des Proteins. Um eine Terti&#xE4;rstruktur zu stabilisieren, interagieren auch Aminos&#xE4;urereste miteinander, die innerhalb der Polypeptidkette weit voneinander entfernt sein k&#xF6;nnen. Diese Wechselwirkungen k&#xF6;nnen schwach und nicht-kovalent (z.B. Ionenbindungen, hydrophobe Wechselwirkungen oder Wasserstoffbr&#xFC;cken) oder stark und kovalent (z.B. Disulfidbr&#xFC;cken) sein. Alle Wechselwirkungen tragen zur Form des Proteins und seiner Funktion bei.</p>
 
 <h4>Mehrere Polypeptidketten k&#xF6;nnen ein einzelnes Protein bilden</h4>
 
 <p>Bisher haben wir Proteine betrachtet, die aus einer einzigen Polypeptidkette bestehen. Viele Proteine bestehen jedoch auch aus Untereinheiten, die jeweils aus einer Polypeptidkette bestehen. Die Zusammensetzung und Interaktion mehrerer Proteinuntereinheiten wird als Quart&#xE4;rstruktur bezeichnet.</p>

<p class="jove_content">Protein organization describes how a protein is arranged at several levels. These levels help explain how a chain of amino acids becomes a working molecule. The main stages are primary, secondary, tertiary, and quaternary structure.</p>

<p class="jove_content">Primary structure is the linear order of amino acids in the protein chain. Secondary structure is the first step of folding. In this stage, the chain can form patterns such as helices and sheets. Tertiary structure is the overall three-dimensional shape of one folded protein chain.</p>

<p class="jove_content">Quaternary structure applies when two or more protein chains join together. Each level adds to the final form and behavior of the protein. Understanding protein organization helps show how structure and function are linked in biology.</p>

Students should address interactions between genes in proteins which regulate both the shape and reproduction of DNA molecules.
