Antibody Structure

Antikörper, auch als Immunglobuline bekannt, spielen eine Schlüsselrolle bei der humoralen Immunreaktion gegen fremde Antigene.
 Es handelt sich um Proteine, die aus vier Polypeptiden in einer Y-förmigen Struktur mit zwei identischen schweren und zwei identischen leichten Ketten bestehen, die durch eine Kombination von nichtkovalenten Bindungen und kovalenten Disulfidbindungen miteinander verbunden sind.
 Ganz am Ende jeder Polypeptidkette befinden sich einzigartige variable Bereiche, die Antigene selektiv binden Während einige Teile dieser Region direkt interagieren, bieten andere Teile eine strukturelle Unterstützung für diese Bindungsstellen.
 Im Gegensatz dazu bestimmt die Stammregion des Antikörpers, die als konstante Region bekannt ist, den spezifischen immunologischen Wirkungsmechanismus in Abhängigkeit von der Klasse.
 Beispielsweise wird ein Typ, Immunglobulin E, als Reaktion auf ein Allergen, wie Katzenschuppen, im Körper ausgeschieden und vermittelt die allergische Reaktion.

 Antikörper, auch Immunglobuline (Ig) genannt, sind wesentliche Akteure des adaptiven Immunsystems. Diese antigenbindenden Proteine werden von B-Zellen hergestellt und machen 20 Prozent des gesamten Gewichts des Blutplasmas aus. In Säugetieren werden Antikörper in fünf verschiedene Klassen eingeteilt, die jeweils eine unterschiedliche biologische Reaktion auf die Antigenbindung auslösen.
 
 <h4>Die Y-förmige Struktur der Antikörpern besteht aus vier Polypeptidketten</h4>
 
 Antikörper bestehen aus vier Polypeptidketten: zwei identischen schweren Ketten mit jeweils etwa 440 Aminosäuren und zwei identischen leichten Ketten mit jeweils etwa 220 Aminosäuren. Diese Ketten sind in einer Y-förmigen Struktur angeordnet, die durch eine Kombination von kovalenten Disulfidbindungen und nicht-kovalenten Bindungen zusammengehalten wird. Darüber hinaus besitzen die meisten Antikörper Zuckerreste. Der Vorgang des Anfügens von Kohlenhydraten an Proteine wird Glykosylierung genannt.
 
 <h4>Die Untereinheiten eines Antikörpers haben unterschiedliche Funktionen</h4>
 
 Sowohl die leichte als auch die schwere Kette tragen zur Antigenbindungsstelle an jeder der Enden der Y-Struktur bei. Diese 110-130 Aminosäuren können sehr variieren, so dass eine fast unbegrenzte Anzahl von Antigenen erkannt werden kann. Diese Region wird auch als variable Region bezeichnet und ist Teil des antigenbindenden Fragments.
 
 Jeder Arm der Y-förmigen Einheit trägt eine identische Antigenbindungsstelle. Antikörper können Antigene miteinander vernetzen, wenn ein Arm an ein Antigen und der andere Arm an ein zweites, strukturell identisches Antigen gebunden werden. Die Quervernetzung wird durch die flexible Gelenkregion (engl. hinge) erleichtert, welche die leichten Ketten des Antikörpers mit dem Stamm verbindet und variable Abstände zwischen den Antigenbindungsstellen ermöglicht. Große Gitter von vernetzten Antigenen werden anschließend schneller und leichter von Makrophagen umschlossen, so dass größere Mengen des Antigens auf einmal beseitigt werden können.
 
 Die Stammregion des Antikörpers wird auch als kristallisierbares Fragment (Fc) bezeichnet und bestimmt die Effektorfunktion des Antikörpers. Über die Fc-Domäne kann der Antikörper mit Fc-Rezeptoren von anderen Immunzellen, wie B-Zellen, Makrophagen und Mastzellen, reagieren. Die Fc-Region ist häufig glykosyliert, wodurch der Zugang des Fc-Rezeptors behindert oder ermöglicht werden kann. Eine Veränderung des Glykosylierungszustandes des Antikörpers erlaubt daher eine schnelle Modulation der Antikörperfunktion.
 
 <h4>Säugetiere verfügen über fünf Klassen von Antikörpern</h4>
 
 Antikörper werden nach der Anzahl der Y-förmigen Strukturen und der Art der schweren Ketten klassifiziert. Antikörper der Klasse IgD, IgE und IgG haben eine einzige Y-förmige Struktur, die zwei identische Antigenbindungsstellen an den Enden der leichten Ketten aufweisen. Wissenschaftlicher ausgedrückt kann man sagen, dass sie eine Wertigkeit von zwei aufweisen. IgD, IgE und IgG unterscheiden sich jedoch in der Zusammensetzung der Disulfid-und nicht-kovalenten Bindungen zwischen ihren beiden schweren Ketten. IgA kann als Monomer oder als Dimer auftreten und ähnelt zwei Ys, die an ihrer Basis verbunden sind. Als Dimer hat IgA vier identische Antigenbindungsstellen mit einer Wertigkeit von vier. IgM kann als Monomer auftreten, wird aber häufiger als Pentamer angetroffen, was ihm eine Wertigkeit von 10 verleiht.
 
 <h4>Die fünf Klassen der Antikörper lösen unterschiedliche Immunreaktionen aus</h4>
 
 IgG-Antikörper sind die am häufigsten im Blut vorkommenden Antikörpermoleküle und werden in großen Mengen ausgeschieden, wenn sich ein spezifischer Erreger ein zweites Mal präsentiert. IgGs tragen in verschiedenen Weisen zur Eliminierung von Krankheitserregern bei. Sie opsonieren Krankheitserreger, um die Phagozytose durch Makrophagen oder Neutrophile auszulösen. Die Aktivität dieser phagozytischen Zellen wird durch das Komplementsystem, eine Kaskade mit enzymatischen Proteinen, verstärkt. Das Komplementsystem selbst wird durch IgG aktiviert. Darüber hinaus sind IgGs die einzigen Antikörper, die durch die Plazenta der Mutter zum Fötus gelangen können. Sie werden auch in die Milch der Mutter sezerniert und bieten so eine passive Immunität, die den Säugling vor Infektionen schützt.
 
 IgA schützt Schleimhautoberflächen wie den Magen-Darm-Trakt, die Atemwege und den Urogenitaltrakt. Es neutralisiert vor allem Bakterien und verhindert ihre Wanderung durch das Epithelium. IgA wird auch in Schleim, Tränen, Speichel und Kolostrum (das antikörperreiche Sekret der mütterlichen Brust in den ersten Tagen nach der Geburt) abgesondert. IgA kommt als Dimer vor, wenn es sekretiert wird und als Monomer, wenn es in Körperflüssigkeiten vorhanden ist.
 
 Monomere der IgM-Klasse sind die ersten, die auf naiven B-Zellen erscheinen. IgMs sind die Hauptklasse von Antikörpern, die von B-Zellen als Reaktion auf die erste Exposition mit einem Antigen sezerniert werden. Sie bilden die primäre Antikörperreaktion. Wenn ein Antigen an ein IgM-Molekül bindet, aktiviert es das Komplementsystem und neutralisiert Krankheitserreger.
 
 Die Funktionen von IgD-Antikörpern sind noch nicht gut verstanden, scheinen aber denen von IgM zu ähneln.
 
 IgE sind aufgrund ihrer geringen Mengen an Körperflüssigkeiten schwierig zu untersuchen und sind vor allem für ihre negativen Auswirkungen auf das menschliche Wohlbefinden bekannt: Allergien. Bei einer allergischen Reaktion bindet das IgE an sein verwandtes Antigen. Anschließend bindet die Fc-Region des IgE an Mastzellen und Basophile, das ist eine Art von weißen Blutkörperchen. Die Interaktion von IgE und dem Fc-Rezeptor auf der Zelloberfläche löst die Freisetzung von Histaminen und Interleukinen aus, was wiederum allergische Symptome wie Niesen und Juckreiz bewirkt.
 
 <h4>Die Antikörper für Forschung, Diagnostik und Therapie werden in Tieren produziert</h4>
 
 In vielen Forschungsdisziplinen sowie bei der Diagnose und auch bei der Behandlung von Krankheiten sind Antikörper ein wichtiges Instrument. Zur Herstellung von Antikörpern wird ein Antigen in ein Nutz -oder Versuchstier injiziert und später wird der Antikörper aus dem Blut des Tieres isoliert. Dabei werden häufig Kaninchen, Hühner, Hamster oder Ziegen verwendet.

Antibodies are built from heavy and light chains that work together to form a Y-shaped molecule. These chains give the antibody its basic structure and help it bind to specific targets.

The two upper arms of the Y contain the variable regions, which differ from one antibody to another. These variable regions help determine which antigen, or foreign molecule, the antibody can recognize.

The lower part of the antibody is the constant region. This region is more similar across antibodies and helps define the antibody class. The structure of the chains also supports the antibody’s overall shape and function.

Biology 1 represents a student's introduction to a molecular model of a DNA as well as the organization of DNA into genes and genes subsequently into chromosomes.
