Ligand Binding and Linkage

Die meisten Proteine haben mehrere Stellen, an denen sich Liganden assoziieren können, zusätzlich zu der Stelle, die für die Proteinfunktion verantwortlich ist. Die Bindung eines Moleküls an eine dieser Stellen führt häufig zu Konformationsänderungen, die die Form des Proteins verändern.
Wenn diese Änderung die Bindung eines anderen Liganden beeinflusst, werden die beiden Stellen als gekoppelt oder verknüpft beschrieben. Die Bindungsstellenkopplung kann positiv oder negativ reguliert werden.
Bei einer positiven Kopplung führt die Bindung eines Liganden zu Konformationsänderungen, die eine Bindung an den jeweiligen Liganden an eine andere Stelle wahrscheinlicher machen. 
Bei einer negativen Kopplung führt die Bindung eines Liganden zu Konformationsänderungen, die verhindern, dass eine andere Bindungsstelle mit ihrem Liganden assoziiert wird.

ligandenbindung und kopplung

Allosterische Proteine besitzen mehr als eine Ligandenbindungsstelle; die Bindung eines Liganden an einer dieser Stellen beeinflusst die Bindung von Liganden an die anderen Stellen. Wenn ein Protein allosterisch ist, werden seine Bindungsstellen als gekoppelt oder verknüpft bezeichnet. Im Fall von Enzymen wird die Stelle, die das Substrat bindet, als aktive Stelle bezeichnet und die andere Stelle als regulatorische Stelle. Wenn ein Ligand an die regulatorische Stelle bindet, führt dies zu Konformationsänderungen im Protein, die die aktive Stelle des Proteins beeinflussen können.

Die Verknüpfung von Bindungsstellen kann entweder eine positive oder negative Regulation der Ligandenbindung an andere Stellen bewirken. In Fällen, in denen beide Liganden die gleiche Konformation eines Proteins bevorzugen, erhöht die Bindung an einer Stelle die Affinität der anderen Stelle für ihren jeweiligen Liganden. Dies wird als positive Verknüpfung bezeichnet. Andererseits, wenn die Liganden verschiedene Konformationen bevorzugen, erschwert die Bindung eines Liganden die Bindung eines zweiten Liganden an das Protein. Dies wird als negative Verknüpfung bezeichnet.

Allosteric proteins have more than one ligand binding site. A ligand is a molecule that binds to a protein. When a ligand binds at one site, it can affect binding at the other sites. These sites are called coupled or linked because they influence one another.

In enzymes, the site that binds the substrate is the active site. The other site is the regulatory site. When a ligand binds to the regulatory site, it can change the protein’s shape. This conformational change can then alter how the active site works.

Linkage between binding sites can be positive or negative. In positive linkage, both ligands prefer the same protein conformation. Binding at one site then increases the affinity, or binding strength, of the other site for its ligand. In negative linkage, the ligands prefer different conformations. Binding of one ligand makes it harder for the second ligand to bind to the protein.

EXPLANATION: Discussions of DNA in earlier grades have been limited to discussions of genes and the role of genes in the appearance and activities of an organism. Biology 1 represents a student's introduction to a molecular model of a DNA as well as the organization of DNA into genes and genes subsequently into chromosomes. Students should address interactions between genes in proteins which regulate both the shape and reproduction of DNA molecules. It is important to note that not all segments of DNA are transcribed for proteins. Portions of DNA are involved in regulating the expression of genes or affect the structure of the chromosomes (replication origin, centromeres, and telomeres).
