Transgenic Plants

Transgene Pflanzen werden hergestellt, indem ein fremdes Gen in das Genom einer Pflanze eingebracht wird. Diese Pflanzen können aus einer Vielzahl von Gründen entwickelt werden, darunter Virus- oder Schädlingsresistenz, höherer Ertrag oder therapeutische Proteinproduktion.
Um eine transgene Pflanze herzustellen, werden ein Gen von Interesse und sein Promotor in den pflanzlichen Protoplasten, eine Pflanzenzelle ohne Zellwand, eingeführt, indem beide in einem Medium inkubiert werden, das Polyethylenglykol enthält.
Der rekombinante Protoplast wird auf einem sterilen Medium in Gegenwart von Wachstumsregulatoren und Nährstoffen kultiviert.
Die Zellen des Protoplasten wachsen unbegrenzt und produzieren eine Masse undifferenzierter Zellen, die als Kallus bezeichnet wird. Einige Zellen des Kallus, die sogenannten totipotenten Zellen, lassen neue Triebe und Wurzeln entstehen, die sich zu einer ganz neuen Pflanze entwickeln können.
Transgene Pflanzen können auch mit dem Bakterium Agrobacterium tumefaciens erzeugt werden, einem gramnegativen Bakterium, das im Boden vorkommt. Dieses Bakterium ist ein Krankheitserreger, der normalerweise Krankheiten in Pflanzen verursacht und ein Ti-Plasmid enthält, das sich in das Genom einer Pflanzenzelle integrieren kann.
Das Ti-Plasmid hat zwei interessante Regionen - T-DNA oder Transfer-DNA-Region, die in das Genom der Wirtszelle eingefügt werden kann, und die Virulenzregion, um den Transfer von T-DNA in die Wirtszelle zu unterstützen.
Dieses Plasmid wird modifiziert, indem ein gewünschtes Gen in die T-DNA-Region und ein Gen für Antibiotikaresistenz eingefügt werden, um ein selektives Wachstum von plasmidhaltigen Organismen zu ermöglichen. Das rekombinante Plasmid wird dann durch Anlegen eines elektrischen Impulses in die Bakterien eingefügt, um die Zellpermeabilität zu erhöhen und die Aufnahme des Plasmids in die Bakterien zu ermöglichen.
Das rekombinante Plasmid Agrobacterium tumefaciens wird in einem antibiotikahaltigen Nährmedium gezüchtet.
Pflanzenteile wie die Keimblattknoten einer Tomatenpflanze werden geschnitten und in das bakterienhaltige Medium inkubiert.
Die verwundeten Pflanzenzellen am Rand des Stammknotens setzen Verbindungen frei, die die Bakterien anlocken. Das Bakterium infiziert die Pflanze und setzt das rekombinante Plasmid frei, das dann das fremde Gen in das Genom der Pflanzenzelle überträgt.
In dem antibiotikahaltigen Medium überleben nur die transformierten Zellen mit rekombinanter DNA und antibiotikaresistenten Genen.
Die transformierten Zellen bilden nun einen Kallus und entwickeln sich zu einer neuen transgenen Pflanze, die ein Transgen trägt.

Die rekombinante DNA-Technologie, die als Transgenese bezeichnet wird, wird häufig verwendet, um ein fremdes Gen hinzuzufügen oder ein schädliches Gen aus einem Organismus zu entfernen. Solche genetisch veränderten Organismen werden als transgene Organismen bezeichnet.

Die erste transgene Pflanze war eine Tabakpflanze, die 1983 entwickelt wurde und Resistenz gegen das Tabakmosaikvirus zeigte. Seitdem wurden viele transgene Pflanzen entwickelt und kommerzialisiert, um den landwirtschaftlichen, zierpflanzlichen und gartenbaulichen Wert einer Nutzpflanze zu verbessern. Transgene Pflanzen können auch als Bioreaktoren zur Produktion wirtschaftlich wichtiger Metaboliten oder Proteine verwendet werden.

Physikalische Methoden zur Übertragung von Genen auf Pflanzen

Viele Methoden, wie Elektroporation und Mikroprojektil-Beschuss, können verwendet werden, um ein fremdes Gen in einen Pflanzenprotoplasten zu übertragen. Bei der Elektroporation liefert ein Gerät Hochspannungsimpulse an die Protoplastensuspension. Dies erhöht die Permeabilität des Protoplasten, sodass die fremde DNA in die Zelle eindringen und sich in das Pflanzengenom integrieren kann.

Im Gegensatz dazu werden beim Mikroprojektil-Beschuss oder der Biolistic-Methode kugelförmige Partikel aus Wolfram oder Gold (Mikroprojektile) mit dem Plasmid, das das interessierende Gen trägt, beschichtet. Die beschichteten Partikel werden dann mit einem Instrument, das als Partikelkanone bekannt ist, auf 300 bis 600 m/s beschleunigt. Das Instrument verwendet hochdruckiges Heliumgas, um die treibende Kraft zu liefern, die hilft, dass die plasmidbeschichteten Partikel die Zellwand der Pflanze durchdringen. Einmal in der Zelle, integriert sich das Gen von Interesse in das Pflanzengenom. Diese Methode kann verwendet werden, um DNA in Kalluskulturen, meristematische Gewebe, unreife Embryonen, Pflanzenzellsuspensionen usw. einzuführen.

Anwendungen transgener Pflanzen

Die rekombinante DNA-Technologie wird weitgehend verwendet, um Pflanzen zu entwickeln, die resistent gegen Schädlinge und Viren sind. Die Technik ist auch hilfreich bei der Modifikation des Lipid-, Stärke- und Proteingehalts der Samen.

Zum Beispiel ist Golden Rice eine genetisch modifizierte Reispflanze mit erhöhtem Nährwert. Diese transgene Reissorte produziert β-Carotin – einen Vorläufer von Vitamin A, das ihr eine goldene Farbe verleiht. Sie wird durch Agrobacterium-vermittelte DNA-Transformation produziert, wobei die rekombinante DNA, die drei Gene kodiert, die Enzyme Phytoensynthase, Phytoendesaturase und Lykopin-β-Cyclase in das Genom der gezielten Reisembryonen eingefügt wird. Die Embryonen entwickeln sich dann zu Pflanzen, die β-Carotin produzieren. Golden Rice wurde entwickelt, um Vitamin-A-Mangel in Entwicklungsländern zu bekämpfen.

Andere transgene Pflanzen, wie Bt-Baumwolle, wurden durch Einfügen eines Gens modifiziert, das von dem Bakterium Bacillus thuringiensis isoliert wurde – ein grampositives Bakterium, das im Boden gefunden wird. Transgenic plants are crops that have been changed with recombinant DNA technology, also called transgenesis. This method adds a foreign gene or removes a harmful one from an organism. The result is a genetically modified organism. The first transgenic plant was a tobacco plant made in 1983, and it was resistant to tobacco mosaic virus.

Since then, transgenic plants have been developed for many uses in agriculture, horticulture, and ornamental plants. Some are designed to resist pests or viruses. Others are used to improve the lipid, starch, or protein content of seeds. In some cases, transgenic plants can also act as bioreactors and make useful metabolites or proteins.

Foreign genes can be transferred into plant protoplasts by physical methods such as electroporation and microprojectile bombardment. In electroporation, high-voltage pulses make the protoplast membrane more permeable. This allows the foreign DNA to enter the cell and integrate into the plant genome. In microprojectile bombardment, also called the biolistic method, tungsten or gold particles are coated with plasmid DNA and shot into the cells with a particle gun.

The particle gun uses high-pressure helium gas to accelerate the coated microprojectiles to about 300 to 600 m/s. The particles can penetrate the plant cell wall and deliver the gene of interest. This method can be used with callus cultures, meristematic tissues, immature embryos, and plant cell suspensions. Once inside the cell, the introduced DNA can integrate into the plant genome.

A well-known example is golden rice, which has enhanced nutritional value. It produces β-carotene, a precursor of vitamin A, and this gives the rice its golden color. Golden rice was made by Agrobacterium-mediated DNA transformation using recombinant DNA with genes for phytoene synthase, phytoene desaturase, and lycopene β-cyclase. It was developed to help address vitamin A deficiency in developing nations.

Another example is Bt cotton. It contains a gene from Bacillus thuringiensis, a gram-positive soil bacterium. The plant makes Cry protein, which kills harmful insects and makes the crop insect resistant. These examples show how transgenic plants can improve crop value and help solve real agricultural problems.
