Ein grundlegender Bestandteil des Lebens ist die Fähigkeit einer Zelle, ihr Genom zu replizieren und sich zu teilen; diese Prozesse finden in zwei Hauptphasen des Zellzyklus statt.
Zunächst wird während der S-Phase die chromosomale DNA dupliziert. Während der M-Phase werden die duplizierten Chromosomen getrennt und auf zwei genetisch identische Tochterzellen verteilt.
Nach der S-Phase ist die DNA der Schwesterchromatiden sehr lang und verworren. Die Trennung der Schwesterchromatiden in diesem Zustand könnte zu Chromosomenbrüchen, unsachgemäßer Segregation und sogar zum Zelltod führen.
Um diese potenzielle Krise abzuwenden, wendet die Zelle während der frühen Mitose eine beträchtliche Menge an Energie auf, um die Schwesterchromatiden allmählich in kürzere Strukturen umzuwandeln, die sich leichter trennen.
Diese Reorganisation beruht auf Condensin, einem Proteinkomplex, der an der Kondensation von Chromosomen beteiligt ist.
Condensin besteht aus fünf Untereinheiten. In Eukaryoten sind die beiden Hauptuntereinheiten, SMC2 und SMC4, an ihren ATPase-Hauptdomänen durch die drei anderen Untereinheiten verbunden: eine Kleisin- und zwei HEAT-repeat-Untereinheiten.
Condensine nutzen die durch ATP-Hydrolyse erzeugte Energie, um zwei wichtige Prozesse zu fördern, die die Trennung der Schwesterchromatiden erleichtern: die Chromosomenkondensation und die Auflösung der Schwesterchromatiden.
Während der Chromosomenkondensation werden die Chromatiden dicht gepackt. Während der Auflösung der Schwesterchromatiden werden die kondensierten Schwesterchromatiden zu unterschiedlichen Strukturen, wenn die Schwester-DNAs durch die teilweise Entfernung von Kohäsinen entkoppelt oder entkatifiziert werden.
Wenn die Zelle die Metaphase erreicht, sind die Schwesterchromatiden entlang der Arme nur noch lose verbunden, aber an den Zentromeren immer noch eng miteinander verbunden. 
Condensin kann das DNA-Coiling verändern und die Chromosomenkondensation und die Auflösung von Schwesterchromatiden katalysieren, indem es ringförmige Strukturen bildet, die DNA-Schleifen umgeben.

Condensins

Condensine sind große Proteinkomplexe, die ATP nutzen, um die Montage von Chromosomen während der Mitose zu fördern. Sie verwandeln die verwickelte, formlose Masse der post-interphasischen DNA in individualisierte Chromosomen, indem sie die chromosomale DNA kompaktieren, organisieren und segregieren.

Pflanzen- und Tierzellen enthalten zwei Typen von Condensin-Komplexen – Condensin I und Condensin II. Beide Komplexe haben fünf Untereinheiten: zwei SMC (Structural Maintenance of Chromosomes) Untereinheiten, eine Kleisin-Untereinheit und zwei HEAT-Repeat-Untereinheiten.

Die Kernuntereinheiten sowohl von Condensin I als auch von Condensin II sind SMC2 und SMC4. SMC-Proteine verändern die Anordnung der DNA in einer ATP-abhängigen Weise. Die anderen drei Untereinheiten – die nicht-SMC- oder Hilfsuntereinheiten – unterscheiden sich zwischen den beiden Komplexen.

Studien, in denen das Wirbeltier-Condensin erschöpft wurde, haben unterschiedliche Rollen für Condensin I und II bei der Bildung mitotischer Chromosomen gezeigt. Die Entfernung von Condensin II führt zu längeren, flexibleren Chromosomen, Chromosomenverwicklungen, sperrigen Chromatinbrücken während der Anaphase und einer drastischen Verkürzung der Prophase. Im Gegensatz dazu führt die Entfernung von Condensin I zu kürzeren, breiteren Chromosomen und einer weniger schwerwiegenden, aber dennoch zur Zytokinese-Fehlschlag führenden Störung der Anaphase.

Eine beliebte Erklärung dafür, wie Condensine Chromosomen kompaktieren, ist das Loop-Extrusionsmodell. Dieses Modell postuliert, dass ein Condensin-Molekül an zwei nahegelegenen DNA-Stellen binden kann und sie in entgegengesetzte Richtungen schiebt, wodurch eine wachsende DNA-Schleife entsteht. Condensine können auch miteinander interagieren, um Multimere zu bilden, die entfernte Segmente des Chromatins verbinden.

Mutationen in Condensin wurden mit mehreren Arten von Krebs in Verbindung gebracht. Zum Beispiel entwickelten Mäuse mit einer Missense-Mutation im Gen für eine Untereinheit von Condensin II T-Zell-Lymphome. Condensins shape chromosomes during mitosis, the stage when a cell divides. These large protein complexes use ATP, the cell’s energy molecule, to help turn loose post-interphase DNA into compact, separate chromosomes. They organize and segregate chromosomal DNA so it can be passed on correctly.

Plant and animal cells contain two condensin complexes: condensin I and condensin II. Each complex has five subunits. Two are SMC, or Structural Maintenance of Chromosomes, subunits. One is a kleisin subunit, and two are HEAT-repeat subunits. The core SMC subunits in both complexes are SMC2 and SMC4, which change DNA arrangement in an ATP-dependent way. The other three subunits are non-SMC, or auxiliary, subunits, and they differ between condensin I and condensin II.

Studies in vertebrate cells show that condensin I and condensin II have distinct roles in mitotic chromosome formation. When condensin II is removed, chromosomes become longer and more flexible. They can also become tangled, form bulky chromatin bridges during anaphase, and move into a much shorter prophase. When condensin I is removed, chromosomes become shorter and wider. The anaphase defect is less severe, but cytokinesis still fails.

One common explanation for chromosome compaction is the loop extrusion model. In this model, a condensin molecule binds two nearby DNA sites and slides them in opposite directions. This creates a growing DNA loop. Condensins may also work together as multimers that connect distant parts of chromatin. Condensin mutations have been linked to several cancers, including T cell lymphomas in mice with a missense mutation in a condensin II subunit gene. Researchers are still working out exactly how condensins control chromosome structure, but these protein complexes are important for the cell cycle and cell survival.
