Die mitotische Spindel trennt die Schwesterchromatiden und bewegt sie während der Anaphase der Mitose auf gegenüberliegende Seiten der Zelle.
Grundlegende Strukturen der mitotischen Spindel sind Hohlzylinder, die als Mikrotubuli bezeichnet werden. Zwei Sätze von Mikrotubuli sind an gegenüberliegenden Enden oder Polen der mitotischen Spindel angeordnet.
Jeder Mikrotubuli hat ein Minus-Ende und ein Plus-Ende. Die Minusenden der Mikrotubuli treffen sich in der Mitte der Spindelpole. Die Plusenden erstrecken sich von den Polen nach außen.
Es gibt verschiedene Arten von Mikrotubuli mit unterschiedlichen Positionen und Rollen in der mitotischen Spindel.
Kinetochor-Mikrotubuli binden Chromosomen an den Spindelpol, indem sie an ihren Pluspunkten an Kinetochore anheften. Kinetochore sind große Proteinkomplexe, die am Chromatidenzentromer assembliert sind, einer spezialisierten DNA-Sequenz, die Schwesterchromatiden verbindet.
Die Anordnung der interpolaren Mikrotubuli ähnelt einem Paar gefalteter Hände. Das Plus-Ende eines interpolaren Mikrotubuli überlappt sich mit dem Plus-Ende eines anderen interpolaren Mikrotubuli, das sich vom gegenüberliegenden Pol erstreckt. Motorproteine assoziieren mit interpolaren Mikrotubuli, um die Spindelassemblierung zu steuern.
Astrale Mikrotubuli verankern die Spindel in der Zelle. Diese Mikrotubuli ähneln zusammen einem Starburst, wobei jedes positive Ende vom Spindelpol nach außen zur Zellrinde ragt.
In den meisten tierischen Zellen sind die Mikrotubuli um eine Organelle herum organisiert, die als Zentrosom bezeichnet wird. An jedem Spindelpol befindet sich ein Zentrosom.
Ein Zentrosom besteht aus zwei Zentriolen, die von einer formlosen Masse von Proteinen umgeben sind, die als perizentrioläre Matrix bezeichnet wird. Das Zentrosom produziert, organisiert und verankert Mikrotubuli in der Zelle.
Zwei Familien von Motorproteinen sind integraler Bestandteil des Aufbaus und des Betriebs der mitotischen Spindel: Kinesin-verwandte Proteine und Dynein. Typischerweise bewegen sich Kinesin-verwandte Proteine zu den Plus-Enden der Mikrotubuli und Dynein zu den Minus-Enden.
Die intrinsische Polarität der mitotischen Spindel und ihrer Mikrotubuli erleichtert die mitotische Segregation der Chromosomen und bereitet die Zelle auf die Teilung vor.

The Mitotic Spindle

Die mitotische Spindel – oder der Spindelapparat – ist eine eukaryontische Zytoskelettstruktur, die aus langen Proteinfasern, sogenannten Mikrotubuli, besteht. Die bei der Zellteilung gebildete Spindel trennt Schwesterchromatiden und bewegt sie zu gegenüberliegenden Enden einer Elternzelle, wo die nun einzelnen Chromosomen auf zwei Tochterzellkerne verteilt werden.

Die bipolare Konfiguration der mitotischen Spindel erleichtert die Chromosomentrennung und bereitet die Zelle auf die Teilung vor. Ein Mechanismus, der die bipolare mitotische Spindelbildung gewährleistet, beruht auf Zentrosomen.

Einige Zellen, wie Eizellen von Wirbeltieren und höhere Pflanzenzellen, haben keine Zentrosomen; Die meisten tierischen Zellen haben jedoch zwei Zentrosomen, wenn sie in die Mitose eintreten. Jedes Zentrosom ist mit einer kreisförmigen Anordnung von Mikrotubuli an gegenüberliegenden Enden – oder Polen – der mitotischen Spindel verbunden. Mit anderen Worten: Zentrosome bilden den Keim für Mikrotubuli.

Motorproteine – insbesondere Kinesine und Dynein – wirken typischerweise an oder in der Nähe der Enden von Mikrotubuli und erleichtern die bipolare mitotische Spindelbildung und die Trennung von Schwesterchromatiden.

Zum Beispiel heften sich Kinesin-5-Motoren in der Mittelzone der Spindel an zwei Mikrotubuli, die von gegenüberliegenden Spindelpolen ausgehen, und schieben sie auseinander. Dieser Prozess fördert die Bipolarität und Dehnung der Spindel, indem er die Spindelpole voneinander wegdrückt.

Es wird angenommen, dass die Kinesin-5-Aktivität durch Kinesin-14 ausgeglichen wird. Kinesin-14-Motoren ziehen an Mikrotubuli, die von entgegengesetzten Polen ausgehen, und bringen die Pole effektiv zusammen. Die koordinierte Aktivität dieser Motoren ermöglicht die korrekte Montage der Spindel.

The mitotic spindle is a microtubule-based structure that helps separate chromosomes during cell division. It forms in eukaryotic cells and moves sister chromatids to opposite ends of the parent cell. This leaves each new cell nucleus with the correct genetic material.

A bipolar spindle has two poles, and that shape is important for chromosome segregation. In many animal cells, two centrosomes are present when mitosis begins. Each centrosome helps organize a circular set of microtubules at one pole, which means centrosomes nucleate microtubules. Some cells, including vertebrate oocytes and higher plant cells, do not have centrosomes.

Motor proteins help build and position the spindle. Kinesins and dynein work at or near microtubule ends and support bipolar spindle formation as well as sister chromatid separation. Kinesin-5 motors at the spindle midzone slide two microtubules apart, which pushes the poles away from each other and helps the spindle elongate.

Kinesin-14 acts in the opposite way. It pulls on microtubules that extend from opposite poles and brings the poles closer together. This balance between kinesin-5 and kinesin-14 helps the spindle assemble correctly. Kinesin-4 and kinesin-10 are chromokinesins, which means they can bind to mitotic chromosomes. They attach to chromosome arms and help push the chromosomes and spindle poles apart.

Dyneins also take part in spindle organization. They link astral microtubules to the actin cytoskeleton and help move the spindle poles away from each other. Together, the spindle structure, its poles, and its motor proteins allow sister chromatids to separate and prepare the cell for proper division.
