In eukaryotischen Zellen bilden Mikrotubuli während der Zellteilung die Hauptbestandteile der mitotischen Spindel und werden für die Chromosomensegregation benötigt.
Mikrotubuli sind extrem dynamisch. Einzelne Mikrotubuli wachsen, schrumpfen und wechseln schnell zwischen der Wachstums- und der Verkürzungsphase. Mikrotubuli weisen eine dynamische Instabilität auf, den unvorhersehbaren Wechsel zwischen Wachstum und Schrumpfung.
Die Verschiebung von Wachstum zu Schrumpfung wird als Katastrophe bezeichnet, und die Verschiebung von Schrumpfung zu Wachstum wird als Rettung bezeichnet. Zu jedem Zeitpunkt setzt sich eine Gruppe von Mikrotubuli aktiv zusammen, während andere sich schnell zerlegen.
Mikrotubuli keimieren und wachsen durch die End-to-End-Polymerisation von GTP-gebundenen Tubulin-Heterodimeren. Das Tubulin-Heterodimer besteht aus einer Alpha- und einer Beta-Untereinheit.
Die Beta-Tubulin-Untereinheit ist an eine hydrolysierbare Form von GTP gebunden. Die Hydrolyse von GTP zu GDP destabilisiert das Mikrotubuli-Gerüst. Die Struktur spreizt sich an der Spitze und der Effekt breitet sich nach unten aus, wodurch die Mikrotubuli depolymerisiert wird.
Eine Vielzahl von regulatorischen Proteinen steuert die Dynamik der Mikrotubuli. Mehrere Mikrotubuli-assoziierte Proteine (MAPs) fördern die Stabilität der Mikrotubuli, während mehrere andere Proteine, die Katastrophenfaktoren, die Mikrotubuli destabilisieren. Zellen verändern die Aktivität regulatorischer Proteine, um die Dynamik der Mikrotubuli in Abhängigkeit von der Phase des Zellzyklus zu verändern.
Zum Beispiel enthalten die meisten tierischen Zellen während der Interphase ein zytoplasmatisches Array aus langen Mikrotubuli, die von einem einzigen Zentrosom ausgehen. Wenn die Zellen in die mitotische Phase übergehen und sich duplizierte Zentrosomen in Richtung der entgegengesetzten Pole bewegen, nimmt die Instabilität der Mikrotubuli zu.
Die Instabilität der Mikrotubuli erleichtert die Bildung einer dichten, dynamischen Anordnung mitotischer Mikrotubuli, die zur Spindelbildung beiträgt.

Microtubule Instability

Mikrotubuli sind hohle zylindrische Filamente mit einem Durchmesser von etwa 25 nm und einer Länge, die von 200 nm bis 25 µm variiert. GTP-gebundene Tubulin-Untereinheiten bilden αβ-Heterodimere für die Mikrotubuli-Montage. Diese grundlegenden Bausteine interagieren längs, polymerisieren zu Protofilamenten. Die Protofilamente interagieren dann durch seitliche Bindungskräfte miteinander, um stabile zylindrische Mikrotubuli zu bilden. Diese zylindrischen Filamente sind dynamisch, da sie wiederholte Montage und Demontage durchlaufen. Diese charakteristische dynamische Instabilität findet sich sowohl in vivo als auch in vitro.

Dynamische Instabilität

Individuelle Mikrotubuli können gleichzeitig an den gegenüberliegenden Enden verlängern und schrumpfen. Ob ein Mikrotubulus wächst oder schrumpft, wird durch seine Raten von Katastrophe und Rettung bestimmt. Katastrophe tritt auf, wenn ein wachsender Mikrotubulus beginnt, sich schnell zu verkürzen. Rettung ist der Übergang eines schrumpfenden Mikrotubulus, um sich schnell zu verlängern. Die Rate der β-Tubulin-gebundenen GTP-Hydrolyse ist ein primärer Faktor, der die dynamische Instabilität bestimmt.

In der Zelle sind sowohl freie Tubulin-Untereinheiten als auch ihre αβ-heterodimeren Formen im zytoplasmatischen Pool vorhanden. Die Polymerisation von Tubulin-Untereinheiten beginnt, wenn GTP-gebundene αβ-heterodimere Untereinheiten eine Schwellenkonzentration überschreiten, die als kritische Konzentration für die Mikrotubuli-Polymerisation bezeichnet wird. β-Tubulin existiert in zwei Formen. Das GTP-gebundene β-Tubulin oder T-Form ist verantwortlich für die Verlängerung und stabile lineare Struktur der Mikrotubuli. Im Gegensatz dazu fördert das GDP-gebundene β-Tubulin oder D-Form den Abbau der Mikrotubuli. Die GTP-gebundenen β-Tubuline am wachsenden Ende wirken als Kappe, die die Krümmung der Protofilamente verhindert und die Verlängerung fördert. Nach der Hydrolyse von GTP wird die Konformation von β-Tubulin leicht verändert, was zur Krümmung der Protofilamente führt. Diese Krümmung erleichtert das Binden von destabilisierenden Proteinen wie Stathmin und Kinesin-13, um die αβ-Tubulin-Heterodimere zu entfernen.

Faktoren, die die Instabilität regulieren

Mikrotubuli-assoziierte Proteine oder MAPs sind entscheidende Regulatoren der dynamischen Instabilität der Mikrotubuli. MAPs werden aufgrund ihrer Funktion in der Mikrotubuli-Dynamik grob als Stabilisatoren und Destabilisatoren klassifiziert. Die stabilisierenden MAPs binden an die Mikrotubuli, um das Katastrophenereignis zu reduzieren und die Verlängerung zu fördern. Andererseits binden die Destabilisatoren, um die Katastrophe zu fördern. Stabilisierende MAPs dominieren während der Interphase und in axonalen und dendritischen Mikrotubuli von Neuronen, die stabile Montagen fördern. Während der Mitose sind destabilisierende MAPs häufiger. Diese MAPs sind verantwortlich für die Chromosomensegregation und den Abbau des zytoskeletalen Netzwerks für die Zellteilung.

Microtubules are hollow, cylindrical filaments in the cell. They are about 25 nm wide and can range from 200 nm to 25 μm in length. Their main building blocks are GTP-bound tubulin subunits that form αβ-heterodimers, which then assemble into protofilaments. These protofilaments join by lateral bonds to create stable microtubule cylinders.

Microtubules are highly dynamic because they repeatedly assemble and break down. They can even grow on one end while shrinking on the other at the same time. This behavior is called dynamic instability. It is seen both in living cells and in vitro.

Whether a microtubule grows or shortens depends on catastrophe and rescue. Catastrophe is the rapid switch from growth to shrinkage. Rescue is the switch from shrinkage back to growth. The rate of GTP hydrolysis on β-tubulin is a major factor that controls this behavior.

Cells contain both free tubulin subunits and αβ-heterodimers in the cytoplasmic pool. Polymerization starts when GTP-bound αβ-heterodimers rise above the critical concentration for microtubule polymerization. β-tubulin also exists in two forms. The GTP-bound T-form supports elongation and a stable, straight microtubule. The GDP-bound D-form favors disassembly.

At the growing end, GTP-bound β-tubulin forms a cap that helps block protofilament bending and supports elongation. When GTP is hydrolyzed, β-tubulin changes shape slightly and protofilaments curve. This curved state helps destabilizing proteins such as stathmin and kinesin-13 bind and remove αβ-tubulin heterodimers.

Microtubule-associated proteins, or MAPs, also regulate dynamic instability. Some MAPs stabilize microtubules, while others promote destabilization. Stabilizer MAPs reduce catastrophe and support elongation. They are common during interphase and in the axonal and dendritic microtubules of neurons, where stable structures are needed. During mitosis, destabilizer MAPs are more common. They help chromosome segregation and the breakdown of the cytoskeletal mesh for cell division.
