Determining the Plane of Cell Division

Eine sich teilende Zelle, die ihre Zellteilungsebene genau bestimmt, ist essentiell für die erfolgreiche Trennung von Chromosomen und die Verteilung des zytoplasmatischen Inhalts an ihre Tochterzellen.
Bei Tieren befindet sich die Teilungsebene am Zelläquator zwischen den segregierten Chromosomen, und ihre Lage wird während der Anaphase bestimmt. Mikrotubuli, die die mitotische Spindel bilden, interagieren mit der Zellrinde und spielen eine entscheidende Rolle bei der Spezifizierung der Spaltfurche.
Drei verschiedene Modelle werden verwendet, um die Bestimmung der Zellteilungsebene zu erklären – das astrale Stimulationsmodell, das polare Relaxationsmodell und das Modell der zentralen Spindelstimulation.
Im astralen Stimulationsmodell interagieren radiale Anordnungen astraler Mikrotubuli mit dem äquatorialen Kortex und induzieren die Bildung der Spaltfurche.
Im polaren Relaxationsmodell senden astrale Mikrotubuli hemmende Signale an die Zellpole und unterdrücken so die Bildung der Spaltfurche an beiden Polen. Dies führt zu einer kortikalen Kontraktion am Zelläquator.
Sobald die Anaphase beginnt, ordnen sich die interpolaren Mikrotubuli, die zwischen den segregierten Chromosomen vorhanden sind, in den überlappenden antiparallelen Mikrotubuli an, die auch als zentrale Spindel bezeichnet werden. Im Modell der Zentralspindelstimulation stimuliert die Zentralspindel die Bildung von Spaltfurchen am äquatorialen Kortex. 
Der allgemeine Konsens ist, dass alle drei Prozesse zusammen wirken, um die Ebene der Zellteilung zu bestimmen. Dieser mehrgleisige Ansatz kann dazu führen, dass das System auf Schwankungen der Proteinkonzentration sowie der Zellgröße reagiert und dadurch die Genauigkeit der Zellteilung erhöht.
Der Beitrag jeder Methode hängt vom Zelltyp und der Zellgröße ab. Zum Beispiel ist in C. elegans-Embryonen ein großes Array von astralen Mikrotubuli vorhanden und könnte daher eine aktivere Rolle bei der Bestimmung der Zellteilungsebene spielen.

Die Positionierung der Zellteilungsebene ist ein kritischer Schritt während der Entwicklung und Zelldifferenzierung, insbesondere während der Mitose, wenn die Ebene entscheidend für die Größe der beiden Tochterzellen ist. Die Zellteilungsebene steht senkrecht zur Ebene der Chromosomensegregation, jedoch haben verschiedene Arten von Organismen unterschiedliche Mechanismen der Zellteilung, die ihrer Morphologie und Funktion entsprechen.

Tierische Zellen

In tierischen Zellen bildet sich die Schnürfurche entlang der Zellteilungsebene, beginnend von der Zellrinde, dem Bereich unterhalb der Plasmamembran. Der mitotische Spindelapparat spielt eine entscheidende Rolle bei der Bestimmung der Zellteilungsebene; daher kann eine Änderung der Achse des mitotischen Spindels die Teilungsebene verändern. Diese Strategie wird von einigen eukaryotischen Organismen wie Caenorhabditis elegans genutzt, um von symmetrischer zu asymmetrischer Zellteilung zu wechseln. Symmetrische Zellteilung produziert zwei identische Tochterzellen und wird oft verwendet, um Gewebe zu wachsen, das viele gleichartige Zellen enthält. Im Gegensatz dazu schafft die asymmetrische Teilung zelluläre Vielfalt, da sie zwei nicht identische Tochterzellen erzeugt. Zum Beispiel vermehren Neuroepithelzellen zunächst ihre Anzahl durch symmetrische Teilungen. Danach nutzen sie asymmetrische Teilung, um Tochterzellen zu produzieren, die der Mutterzelle ähnlich sind sowie differenzierende Neuronen.

Hefe

In Sprosshefe, wie Saccharomyces cerevisiae, wird die Ebene der Zellteilung in der G1-Phase durch Septin-Proteine bestimmt. Die Zytokinese wird durch die Bildung einer Schnürfurche im Halsbereich der Sprosshefezellen erreicht; jedoch spielt der mitotische Apparat keine Rolle bei der Bestimmung der Zellteilungsebene. Im Gegensatz dazu entscheidet in der Spalthefe, wie Schizosaccharomyces pombe, die Position des Interphasekerns in der G2-Phase über den Ort der Zellteilungsebene. Während der Interphase ist das Mid1-Protein hauptsächlich im Kern lokalisiert; jedoch wird es während der Mitose aus dem Kern exportiert und rekrutiert im Zellkortex Komponenten des Aktomyosinrings in den mittleren Bereich der Zelle. Dies führt letztlich dazu, dass sich die Mutterzelle in zwei gleich große Tochterzellen teilt.

Pflanzen

In den meisten Pflanzenzellen wird die Ebene der Zellteilung vor dem Eintritt in die Mitose festgelegt. Ein Präprophaseband bildet sich entlang des Äquators der Elternzelle, das die Zellteilungsebene markiert. Das Präprophaseband ist eine ringförmige Struktur aus Mikrotubuli und F-Aktin. Es gibt Anlass zu einem Mikrotubuli-Bündel namens Phragmoplast, das ein strukturelles Gerüst für die Expansion und Positionierung der Zellplatte bietet.

How cells set the division plane during cell division affects daughter cell size and, in many cases, cell fate. The plane of division is perpendicular to the plane of chromosome segregation. Different organisms use different cues to place it, based on their shape and needs.

In animal cells, the cleavage furrow forms along the division plane from the cell cortex, the area just below the plasma membrane. The mitotic spindle is the main guide for this position. If the spindle axis changes, the division plane can change as well. Some eukaryotes, such as Caenorhabditis elegans, use this shift to move from symmetric division to asymmetric division.

Symmetric cell division makes two identical daughter cells and is often used to build tissues with many of the same cell type. Asymmetric division makes two different daughter cells and increases cell diversity. Neuroepithelial cells first divide symmetrically to raise their numbers. They later divide asymmetrically to form a daughter cell like the parent cell and another that will become a neuron.

In budding yeast, such as Saccharomyces cerevisiae, septin proteins decide the plane of division during the G1 phase. The cell then completes cytokinesis by forming a cleavage furrow at the bud neck. In this yeast, the mitotic apparatus does not set the division plane.

In fission yeast, such as Schizosaccharomyces pombe, the interphase nucleus helps decide where the division plane will form during the G2 phase. The Mid1 protein stays mainly in the nucleus during interphase. During mitosis, Mid1 moves to the cell cortex and recruits actomyosin ring components to the middle of the cell. This lets the mother cell divide into two similar-sized daughter cells.

In most plant cells, the division plane is set before the cell enters mitosis. A preprophase band forms along the cell equator and marks where division will occur. This ring-like structure contains microtubules and F-actin. It helps form the phragmoplast, which supports cell plate growth and placement.
